摘要
以马铃薯块茎为材料,通过差速离心分离出线粒体,过垫梯度离心进一步纯化。显微镜下观察所得的线粒体的纯度、形态;并用数码相机进行照相。线粒体样用SDS、蛋白酶K、核酸酶处理后用饱和酚、苯酚/氯仿及氯仿/异戊醇提取处理,乙醇沉淀,提取得线粒体DNA(mtDNA)。紫外分光光度计检测提取浓度、纯度,1%琼脂糖电泳获得清晰、整齐的条带。
Extract the mitochondrion from the potato. Separate by the differential centrifugation and pass gradien pad centrifugation. Observe pure and shape of the mitochondria from microscope and take pictures with digital camera. Then handle them with SDS,DNaseI and proteinase K. After that we extract the mtDNA (mitochondrion DNA)with phenol/chloroform and chloroform/isoamyl alcohol precipitant. Examine with extract with the 1% agarose gel electrophoresis and quantity of the mtDNA with UV-spectrophotometer. Result showed that the band of mtDNA is very regular.
