乙醛脱氢酶高产菌株Z07-J01的选育与酶学特性 被引量:10

摘要 通过紫外线和He-Ne激光诱变醋酸菌,选育出1株乙醛脱氢酶高产菌株Z07-J01,该正突变株24 h发酵酶活为1 008.00 U/g湿菌体,比出发株提高了276%,且酶活在10代内稳定。该突变株所产乙醛脱氢酶的最适作用pH为7,最适作用温度为50℃;在25℃保温30 min,该酶酶活不受影响;在65℃保温30 min,该酶完全失活;2 mmol/L Cu2+能使该酶完全失活,2 mmol/L Mn2+则对该酶有较强的激活作用。 In this experiment, one Acetobacter mutant Z07-J01 producing more aldehyde dehydrogenase (ALDH) was obtained by Ultraviolet radiation and He-Ne laser . The enzyme activity was 1008.00 U/g wet acetobacter after 24 h incubation, enhanced by 276 %. The enzyme activity of Z07- J01 would be stable after tenth generation. The optimized pH of ALDH was 7 and the optimized temperature was 50℃. The enzyme activity would not be influenced after the ALDH was kept for 30 minutes under 25℃ ,but under 65℃ for 30 minutes, the enzyme would have no activity. 2 mmol/L Cu^2+ would inhibit the enzyme completely while 2 mmol/L Mn^2+ enhances its activity strongly.
