摘要
通过单因素实验和正交实验,对MHL4.07菌摇瓶发酵产脂肪酶的培养基组成和培养条件进行了研究,结果表明:最佳发酵培养基配方为橄榄油15.0g/L、玉米浆40.0g/L、NH4Cl20.0g/L、豆饼粉20.0g/L、MgSO40.5g/L、KH2PO410.0g/L、KCl1.0g/L。最佳发酵条件为种子培养84h、接种量7%、初始pH6.5、装液量50mL/250mL三角瓶,培养温度28℃,摇床转速160r/min,发酵培养96h。在此条件下,最高脂肪酶产量达到60.54U/mL,比优化前提高了44.5%。
The influences of fermentation conditions on lipase produced by Rhizopus arrhizus were investigated through the single factor and the orthogonal experiments. The results show that: The compositions of optimal fermentation medium were olive oil 15.0g/L, corn steep liquor 40.0g/L, NH4Cl 20.0g/L, bean cake flour 20.0g/L, MgSO4 0.5g/L, KH2PO4 10.0g/L, KCl 1.0g/L. The optimum conditions were: the culture time of seed 84h, seed volume 7%, the initial pH 6.5, 50mL in 250mL total volume, the temperature of the shaking table 28%, rotating speed 160r/min, the fermentation time 96h. The maximum yield of lipase was 60.54U/mL and it was increased 44.5% under the optimum conditions.
