摘要
采用单因素试验和正交试验对筛选出的1株生防拮抗菌——枯草芽孢杆菌(Bacillussubtilis)的液体发酵培养基和工艺条件进行优化。确定了最佳的培养基配方为:蔗糖20g·L-1、硫酸铵5g·L-1、麸皮50g·L-1、柠檬酸三钠2.5g·L-1、K2HPO·43H2O0.3g·L-1、MgSO4·7H2O0.5g·L-1、FeSO4·7H2O0.05g·L-1;最适发酵条件为:发酵温度30℃,初始pH值7.2,最适接种量5%;进行了100L发酵罐放大培养,20h达到生长高峰期,生长周期24h,最适放罐时间为26 ̄28h;此时所获得的菌体数量约为6×109个·mL-1。
Bacillus subtilis is potential to play an important biological control plant pathogens effect. The liquid fermentation medium and technological conditions of Bacillus subtilis were optimized by single factor experiments and orthogonal experiments. The results showed that optimum medium formula was sucrose 20 g·L^-1, (NH4)2SO4 5 g·L^-1, wheat bran 50 g·L^-1, citrate sodium 2.5 g·L^-1, KH2PO4.3H,O 0.3g·L^-1, MgSO4^*7H2O 0.5g·L^-1, FeSO4^*7H2O 0.05 g·L^-1. The favorable fermentation condition was determined: fermentation temperature 30 OE, initial pH value of medium 7.2, inoculation amount 5%. The magnifying culture in 100 L fermentation tank was conducted, growth peak value appeared in 20 h, growth period 24 h, and optimum cultivating time 26-28 h, and the obtained bacterium amount was up to 6×10^9 ·mL^-1 in fermentation liquid.
