摘要
目的:探讨衰老雄性大鼠睾丸组织抗氧化酶水平对血清睾酮(T)水平、睾丸间质细胞凋亡蛋白酶-3(caspase-3)基因表达的关系。方法:SD大鼠(20只)随机均分为对照(C)组和D-半乳糖(D)组,用分光光度计检测睾丸组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的含量,放免法检测血清中T水平以及免疫组化检测睾丸间质细胞caspase-3基因表达情况。结果:①SOD活性:D组[(116.0±18.1)NU/mgprot]显著低于C组[(156.0±31.0)NU/mgprot](P<0.01);②GSH-Px水平:D组[(29.8±3.8)U/mgprot]显著低于C组[(35.3±4.5)0U/mgprot](P<0.05);③血清T:D组[(0.7±0.3)ng/mL]显著低于C组[(2.6±0.7)ng/mL](P<0.01);④caspase-3表达:D组[(11.3±1.8)%]明显高于C组[(9.1±1.3)%](P<0.01)。结论:衰老大鼠睾丸组织抗氧化酶活性下降与血清T水平和睾丸间质细胞caspase-3基因表达增强有关。
Objective: To evaluate the effect of antioxidase to serum testosterone(T)and caspase-3 expression in Leydig cell of aging male rats. Methods: Twenty SD rats were randomly divided into two groups with ten rats each: the D-galactose group(D group)and the control group(C group). The levels of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px) in testis were determined by the method of spectro-absorptiometer and the expression of caspase-3 in Leydig cell was observed by immunohistochemical. The concentration of serum T was examined by radio-immunity technique. Results: As to the activities of SOD in testis: D group[(116.0 ± 18.1)NU/mgprot]was statistically lower than C group[(156.0± 31.0)NU/mgprot] (P 〈 0.01). The activities of GSH-Px in testis: D group[ (29.8 ± 3.8)U/mgprot] was statistically lower than C group[ (35.3 ±4.5)U/mgprot] (P 〈 0.05). Serum T in D group[ (0.7 ± 0.3)ng/mL]was markedly lower than that in C group[ (2.6 ± 0.7)ng/mL] (P 〈 0.01 ). The expression of caspase-3 in Leydig cell of D group[ ( 11.3 ± 1.8) % ] was remarkly more than C group [ (9.1 ± 1.3 ) % ] ( P 〈 0.01 ). Conclusion: Lipid peroxidation is concerned with the concentration of serum T and the expression of caspase-3 in Leydig cell of aging male rat.
