摘要
目的探讨内消连翘丸对体外培养的人结节性甲状腺肿细胞(结甲细胞)增殖及分泌的影响。方法分离并培养人的原代结甲细胞,同时用内消连翘丸或优甲乐的含药动物血清进行干预,采用MTT法和放免法检测含药血清对结甲细胞增殖及激素分泌功能的影响。结果各组含药血清对体外培养的结甲细胞的增殖均有明显的抑制作用,并呈一定量效和时效关系,其中内消连翘丸大、中剂量组优于优甲乐组。促甲状腺素(TSH)对结甲细胞的增殖和分泌功能均有促进作用,中药各组含药血清对加TSH刺激的结甲细胞增殖仍有不同程度的抑制作用,中药作用24 h对结甲细胞的激素分泌功能无明显影响。结论在一定时间范围内,内消连翘丸合药动物血清可在不明显影响细胞分泌功能的前提下抑制结甲细胞的增殖。
Objective To investigate the influence of Neixiaolianqiao Pills on the proliferation and secretion of human nodular goiter cells cultured in vitro. Methods The human primary nodular goiter cells were separated and cultured, and then interfered with animal serum containing Neixiaolianqiao Pills or Euthyrox. MTT method and radioimmunoassay were used to detect the influence of the medicated serum on the proliferation and hormone secretion of nodular goiter cells. Result The medicated serum had obvious inhibitory effect on the proliferation of nodular goiter cells cultured in vitro in different groups and showed a dose-effective relation. The results of the groups with high or medium dose of Neixiaolianqiao Pills were better than Euthyrox group. Thyroid-stimulating hormone (TSH) has the promoting effect on the proliferation and secretion of human nodular goiter cells, while the medicated serum had the inhibitory effect on the proliferation of human nodular goiter cells stimulated by TSH in varying degrees in different Neixiaolianqiao Pills groups. Neixiaolianqiao Pills has no significant influence on hormone secretion of human nodular goiter cells 24 hours after its administration. Conclusion The serum containing Neixiaolianqiao Pills can inhibit the proliferation of human nodular goiter cells without significant influence on hormone secretion within a certain period.
