摘要
目的观察中药丹参与卡铂联合应用于胃癌腹腔化疗的疗效。方法利用Walker-256肿瘤细胞株复制成功Wistar大鼠种植性胃肿瘤模型,随机分为对照组(生理盐水组)、卡铂组及丹参+卡铂组,每组20只,于种植成功后1周分别给予生理盐水、卡铂及卡铂+丹参腹腔注射,每日1次,连用5 d。于化疗结束后第7天以颈椎脱位法每组各处死10只,剥离原位肿瘤,测量肿瘤的最大径和最小径,计算肿瘤体积抑制率;将剥离的原位肿瘤以10%中性甲醛固定后24 h内予以常规石蜡包埋后切片,TUNEL法检测细胞凋亡,IMS细胞图像分析系统自动测量凋亡细胞面积及光密度。余下每组10只大鼠,继续相同条件下饲养,观察各组动物的生存时间,并计算出生存时间延长率。结果腹腔化疗结束后1周,给药组肿瘤体积明显缩小,卡铂+丹参组肿瘤体积较卡铂组小;给药组凋亡面积、凋亡强度均增大,卡铂+丹参组肿瘤凋亡面积、凋亡强度较卡铂组明显增大;给药组的中位生存期明显长于对照组,但卡铂组与丹参组的中位生存期无统计学差异。结论卡铂腹腔化疗时联合应用丹参可以增强卡铂诱导肿瘤细胞凋亡的作用以及增加凋亡强度。
Objective It is to observe the curative effect of intraperitoneal chemotherapy (IPC) with combined administration of radix salviae mihiorrhizae (RSM) and Carboplatin on gastric tumor. Methods 60 rat models of planted gastric tumor were made with malignant tumor cell line, and randomly divided into control group, Carboplatin group, RSM & Carboplatin group, and given normal saline, Carboplatin and RSM & Carboplatin respectively by intraperitoneal injection once a day and continually for 5 days in one week after successful planting. 10 rats in every group were killed with cervical vertebra luxation method on the 7th day after the ending therapy to peel off original tumor. The volume of the tumor was measured, and tumor control ratio was calculated. Then the tun:or was fixed with 10 % neutral formaldehyde and made into section. Apoptosis of the cells was detected by TUNEL, apoptosis area and optical density was measured by IMS cell image analyze system. The rest rats in every group were fed in the same condition until they died. The survival time was observed and extend rate of the time was calculated. Results After a week's intraperitoneal chemotherapy, tumor volume was significantly lower and tumor apoptosis area and optical density were significantly bigger in Carboplatin group, RSM & Carboplatin group than those in control group. Compared with each other, the same result can be found in Carboplatin group and RSM & Carboplatin group. Median survival time in Carboplatin group and RSM & Carboplatin group was significantly longer than that in control group, but there was no statistically significance between the former two groups. Conclusion With combined administration of RSM in IPC can enhance Carboplatin's induce function on apoptosis of tumor cells and increase the degree of anti-tumor.
