摘要
对分离自流沙河流域不同海拔高度土壤中的53株自生固氮菌,进行了数值分类及BOX-PCR分析。数值分析结果表明,供试菌株在碳、氮源的利用及抗逆性能等方面存在明显差异,表现出丰富的表型多样性,在82.6%相似水平处,所有供试菌株被分成17个表观群,其中第5和第12表观群最大,分别由9个和14个菌株组成,表明它们是该区域的优势自生固氮菌;BOX-PCR较好地揭示了不同菌株间的差异,供试菌株被划分成10个不同的遗传类群,聚群结果与数值分类有较好的一致性。
53 free-living Dizotrophs isolated from soils at different altitude in Liusha fiver valley were analyzed by phenotypic features and BOX-PCR fingerprinting procedure. The results showed that the strains differed in physiological and biochemical characteristics, including carbon and nitrogen sources, antibiotic resistance, etc. Dendrogram of numerical taxonomy showed that all the tested strains were divided into 17 independent groups at the similarity of 82.6%; group 5 and group 12 were the two largest among these groups, and were constructed by 9 and 14 strains, respectively, suggesting that strains in these two groups were the dominant ones in these areas. Analysis of BOX-PCR also revealed the genetic diversity of the strains properly, and all the strains clustered into 10 different genetic groups. The results of BOX-PCR were correlated well with numerical analysis.
