昆明山海棠治疗耳胆脂瘤的实验研究 被引量:1

摘要 目的应用昆明山海棠对耳胆脂瘤动物模型治疗后,进行TGF-β1、C-FOS在耳胆脂瘤上皮中的表达的研究,探讨昆明山海棠在局部治疗耳胆脂瘤方面的价值及应用原理.方法制作耳胆脂瘤动物模型,以氯霉素、昆明山海棠、生理盐水制剂,进行局部对照治疗,应用免疫组化SP染色法检测TGF-β1、C-FOS分别在以上3组动物模型胆脂瘤上皮中、17例对照健康耳中的表达,应用计算机图像分析系统对其阳性表达进行定量分析.结果应用昆明山海棠、氯霉素局部治疗后的动物模型,其耳胆脂瘤C-FOS的表达明显低于正常外耳道上皮组织(F=7.293,P<0.05),而仅有昆明山海棠制剂使其TGF-β1表达增强(F=5.789,P<0.05),同时也发现生理盐水对TGF-β1、C-FOS的表达无影响.结论即刻早期基因C-FOS在胆脂瘤中表达显著增强,并与胆脂瘤侵袭能力的高低相关.而TGF-β1名为生长抑制多态因子,当在胆脂瘤中高表达使其抑制细胞生长,使胆脂瘤的高增殖处于可控制状态.提示应用昆明山海棠制剂通过细胞因子作用于耳胆脂瘤动物模型,可达到治疗胆脂瘤的目的. Objective To investigate the role of triptergium hypoglaucum on cholesteatome and the expression of C - FOS and TGF -β1 in cholesteatome epithelium during treating ear cholesteatome of the menggu mouse. Method We applied chloramphenicol eye drops, tripterygium hypoglaucum or 0. 9% liquid of the sodium chloride to treat the ear cholesteatome of the animal model ( Menggu mouse) which have been established. By the local therapy, we detected the expressing of C - FOS and TGF -β1 in cholesteatome using Immunohistochemical staining ( SP method) . The expression of them was observed under microscope, after that, we scanned it by an image scanner and quantified the gray of them using commercial software. Result The expression of C - FOS on epithelium of the normal external auditory meatus was notably higher than that on cholesteatome epithelium of the sick ear, which was treated by triptergium hypoglaucum and chloramphenicol eye drops ( F = 7. 293, P 〈0. 05 ) . What's more, the expression of TGF -β1 was higher only on triptergium hypoglaucum treating team ( F = 5.789, P 〈 0. 05 ). In the meantime , we detected that 0. 9% liquid of the sodium chloride could not effect the expression of the C - FOS and TGF -β1. Conclusion It had been definite that the expression of the C - FOS on cholesteatome was higher on the normal epithelium, TGF -β1, growth restrain factor, could restrain and control the growth of cell and its high proliferation. So it was conclude that Triptergium hypoglaucum could play a important role in treating ear choleasteatome of animal model through cell factors such as C - FOS and TGF-β1.
