人参皂苷Rb_1与芳维甲酸乙酯对光老化皮肤真皮成纤维细胞基质金属蛋白酶影响的比较研究 被引量:14

摘要 目的通过人参皂苷Rb1与芳维甲酸乙酯对皮肤真皮体外培养光老化成纤维细胞胞质内基质金属蛋白酶表达的影响,阐明人参皂苷Rb1对光老化的防治机理,探索光老化的防治手段。方法建立体外培养真皮成纤维细胞光老化模型,用人参皂苷Rb1与芳维甲酸乙酯进行干扰,免疫组织化学技术显示基质金属蛋白酶的表达的改变。结果人参皂苷Rb1与芳维甲酸乙酯对培养成纤维细胞胞质基质金属蛋白酶的表达有显著抑制作用(P<0.05);人参皂苷Rb1和芳维甲酸乙酯对其表达的抑制作用相互之间差异无显著性(P>0.05)。结论人参皂苷Rb1对光老化成纤维细胞具有与芳维甲酸乙酯相似的防治作用,其机理与芳维甲酸乙酯的作用机理可能相同或者相似,及通过调节MMPs活性防治皮肤光老化。 Objective To explore the methods of prevention and cure of sun - induced aging dermis, and clarify the mechanism of Ginsenoside Rb1 on sun - induced aging dennis, with the study of expression of matrix metallo preteinases in sun - induced aging dermis fibroblasts'cytoplast, which are cultured in vitro, under the effect of Ginsenoside Rb1 and arotinoid ethyl ester. Methods Build the dermis fibroblasts sun -induced aging model, and affect the cells with Ginsenoside Rb1 and arotinoid ethyl ester, and display the change of expression of matrix metallo preteinases with immunohistochemistry. Results Ginsenoside Rb1 and arotinoid ethyl ester had significant inhibition effect on the expression of matrix metallo preteinases in the fibroblasts'cytoplast cultured in vitro( P 〈 0.05) ; the inhibition roles of Ginsenoside Rb1 and arotinoid ethyl ester on the expression of matrix metallo preteinases were not significant( P 〉 0.05). Conclusion Ginsenoside Rb1 has effect of prevention and cure of sun - induced aging dermis 's fibroblasts like the arotinoid ethyl ester, and its mechanism is the same as or like the arotinoid ethyl ester through regulating the expression of matrix metallo preteinases.
