扶正排毒片对氢化可的松致免疫抑制小鼠免疫功能的调节 被引量:7

摘要 目的:观察扶正排毒片对氢化可的松所致的免疫抑制小鼠免疫功能的影响,探讨扶正排毒片的益气滋阴,清热解毒的功效。方法:实验于2005-02/10在河南中医学院动物实验中心完成。实验分为3批,每批选用小鼠60只,随机数字表法分为6组,每组10只,其中5组皮下注射氢化可的松制备免疫抑制小鼠模型。于造模第1天,高、中、低剂量扶正排毒片组分别灌服0.15,0.10,0.05g/mL扶正排毒片(主要由西洋参64.0g,黄芪160.0g,连翘106.7g,白花蛇舌草160.0g,甘草64.0g等药组成,河南中医学院第三附属医院)混悬液(200mL/kg),香菇多糖片组灌服5g/L香菇多糖片混悬液(200mL/kg),模型组灌服同体积的生理盐水。另有一组为空白对照组,每鼠仅灌服同体积的生理盐水。每天给药1次,连续给药7d。①实验1:第1批小鼠于第7天早上每鼠均腹腔注射5%鸡红细胞液0.5mL,于给鸡红细胞2h后给药,给药后2h,即给鸡红细胞后4h处死小鼠,脱颈椎处死小鼠。观察小鼠腹腔巨噬细胞吞噬情况。②实验2:第2批小鼠于给药第1天每鼠腹腔注射5%鸡红细胞液0.2mL,于最后一次给药后2h,小鼠眼眶取血,分离血清。取上清液于UV-2000型分光光度计540nm处比色。处死小鼠后,取脾细胞混悬液,取上清液于UV-2000型分光光度计413nm处比色。观察溶血素形成和空斑形成细胞情况。③实验3:第3批小鼠于给药第1,2,3天每鼠加肌注0.8g/L植物血凝素(8mg/kg),于最后一次给药后2h,小鼠剪尾取血,推片,瑞氏染色,油镜观察,计算外周血淋巴细胞转化百分率。结果:纳入小鼠180只,均进入结果分析。①实验1:高、中、低剂量扶正排毒片组和香菇多糖片组腹腔巨噬细胞吞噬百分率和吞噬指数高于模型组[分别为(52.8±7.6)%,(55.6±7.2)%,(45.9±7.1)%,(48.7±8.3)%,(35.3±7.1)%,P<0.01;0.82±0.08,0.84±0.07,0.66±0.10,0.67±0.11,0.55±0.10,P<0.05,0.01]。②实验2:高、中、低剂量扶正排毒片组和香菇多糖片组小鼠溶血素形成和空斑形成细胞(A)高于模型组(分别为0.120±0.015,0.131±0.019,0.101±0.017,0.104±0.018,0.063±0.020;0.446±0.047,0.498±0.016,0.383±0.060,0.387±0.064,0.221±0.061,P<0.01)。③实验3:高、中、低剂量扶正排毒片组和香菇多糖片组小鼠外周血淋巴细胞转化率高于模型组[分别为(60.9±5.0)%,(54.8±6.7)%,(49.3±5.9)%,(51.9±10.4)%,(32.5±4.5)%,P<0.01]。结论:扶正排毒片可显著促进氢化可的松所致免疫抑制小鼠腹腔巨噬细胞的吞噬功能,显著促进溶血素形成和空斑形成细胞,显著促进外周血淋巴细胞转化。 AIM: To observe the influence of Fuzheng Paidu tablet on the immunological function of immunosuppressional mice model caused by injecting hydrocortisone, and confer the effect of Fuzheng Paidu tablet on supplementing qi, nourishing yin and expelling toxin by cooling. METHODS: The experiment was accomplished at Animal Experiment Center of Henan College of Traditional Chinese Medicine from February to October in 2005. The experiment was disparted into 3 parts, and 60 mice in every part were randomly divided into 6 groups, with 10 mice in each. Five groups of them were induced to establish immunosuppressional mice models by the subcutaneous injection of hydrocortisone. On the first day of modeling, the large, middle and small-dose Fuzheng Paidu tablet groups were treated with 200 mL/kg suspension containing 0.15, 0.10 and 0.05 g/mL tablets (the mostly ingredient were 64.0 g Panax quinquefolium L., 160.0 g Astragalus membranaceus Bge, 106.7 g Forsythia suspense Vahl, 160.0 g Oldenlondia diffusa Roxb and 64.0 g Glycyrrhiza uralensis Fisch, and the medicine was made by the Third Affiliated Hospital of Henan College of Traditional Chinese Medicine). Xianggu Duotang tablet group were administrated with 200 mL/kg suspension containing 5 g/L tablet, while both model group and control group received the same eubage of physiological saline. Medicines were given once a day for 7 successive days. ①Experiment 1: On the morning of the 7^th day, every mice were injected with 0.5 mL chicken red blood cell liquid (5%) in abdominal cavity, which was followed by administrations 2 hours later. And another 2 hours later, mice were executed for observing the phagocytosis of macrophages in abdominal cavity,②Experiment 2: At the first day morning, every mice was injected with 0.2 mL chicken red blood cell solution (5%) in abdominal cavity. Two hours after the last time of giving the medicine, blood sample was adopted from the mice's eyepit for separating the serum. The level of hemolysin in blood serum was determined by using the spectrophotometer UV-2000 (k=540 nm). Then the mice were executed to break out the spleen suspension. The level of hemolysin and plaque forming cell were observed by using the spectrophotometer UV-2000 (λ =413 nm). ③Experiment 3: On the 1^st, 2^nd and 3^rd days, the mice were injected with 8 mg/kg PHA (0.8 g/L) by intramuscular injection. Two hours after the last medicine, hlood sample was adopted by cutting the mice tails, and the percent of the transformation efficiency of lymphocyte in peripheral blood was counted, RESULTS: All 180 animals entered into the result analyzing. ①Experiment h Compared with the model group, the large, middle and smll-dose Fuszheng, Paidu tablet and Xianggu Duotang tablet remarkably promoted the rate and index of phagocytosis of macrophage in abdominal cavity [(52.8±7.6)%, (55.6±7.2)%, (45.9±-7.1)%, (48.7±8.3)%, (35.3±7.1)%, P 〈 0.01; 0.82±0.08, 0.84±0.07, 0.66±0.10, 0.67±0.11, 0.55±0.10, P 〈 0.05, 0.01],② Experiment 2: Compared with the model group, the large, middle and small-dose Fuzheng Paidu tablet and Xianggu Duotang tablet remarkably improved the level of hemolysin and plaque forming cell in blood serum (0.120±0.015, 0.131±0.019, 0.101±0.017. 0.104±0.018, 0.063±0.020; 0.446±0.047, 0.498±0.016, 0.383±0.060, 0.387±0.064, 0.221 ±0.061. P 〈 0.01).③Experiment 3: Compared with the model group, the large, middle and small-dose Fuzheng Paidu tablet and Xianggu Duotang tablet remarkably increased the transformation efficiency of lymphocyte of the immunosuppressional mice [(60.9±5.0)%, (54.8±6.7)%, (49.3±5.9)%, (51.9±10.4)%, (32.5±4.5)%, P 〈 0.01]. CONCLUSION: Fuzheng Paidu tablet can remarkably accelerate the phagocytosis of macrophage in abdominal cavity, the level of hemolysin and plaque forming cell in blood serum and the transformation of peripheral lymphocyte in immunosuppressional mice.
