摘要
目的探讨应用TGF-β_1多克隆抗体抑制术后屈指肌腱粘连的效果。方法将108只来亨鸡随机分成3组,行中趾屈指肌腱切断吻合术,术中分别滴加生理盐水或低、高浓度的TGF-β_1多克隆抗体,于术后1、3、8、12周取肌腱进行生物力学测试、苏木精-伊红染色、Masson胶原染色、苦味酸-天狼星红染色、TGF-β_1免疫组化染色等。结果TGF-β_1多克隆抗体药物组(5μg/ml组、10μg/ml组)肌腱最大延伸率、模拟主动屈曲度要高于对照组,肌腱最大抗拉力在8周时低于对照组,在12周时与对照组无差异;与对照组比较,10μg/ml组炎症过程缩短,塑形过程加速;药物组(5μg/ml组、10μg/ ml组)Ⅰ胶原含量减少,Ⅰ/Ⅲ胶原比例下降,TGF-β_1的表达降低。结论TGF-β_1多克隆抗体可以有效的抑制TGF-β_1在肌腱损伤修复过程中的作用,减少肌腱的粘连,瘢痕组织的形成,而不影响肌腱的强度,这可能是屈肌腱损伤术后粘连潜在的有效方法之一。
Objective To investigate the preventive effect of TGF-β1 neutralizing antibody on flexor tendon adhesion from operation. Methods One hundred and eight leghon cocks performed anastomonsis operation were divieded into three groups randomly, as normal saline( control group), 5 μg/ml group, 10 μg/ml group of TGF-β1 antibody. At 1 st, 3rd, 8th and 12th weeks respectively after operation, the flexor biomechanics test, HE staining, Masson staining, Sirius red-polarization staining and TGF-β1 immunohistochemistry stai- ning were used. Results The max of strength of tendon and the stimulate active flexor from the experiment groups(5 μg/ml group, 10 μg/ml)are higher than from the control group, The max of strength of tendon of the experiment groups are less at 8th weeks, and no difference at 12th weeks from the control group; Compared with the control group, the 10 μg/ml group were less shorten the progress of inflammation and accelerated the progress of molding; In the experiment groups(5 μg/ml group, 10 μg/ml) ,the density of the collagen Ⅰ type were less, the ratio of Ⅰ /Ⅲ collagen and expression of the TGF-β1 were decreasing. Conclusion The study showed that applying of TGF-β1 multiclonal neutralizing antibody can inhibit efficiently the function of the TGF-β1 during the flexor tendon repair, reduce tendon adhesion and scar fromation, however has no affection of tendon intensity, suggesting it is a latent and efficient method for preventiong flexor tendon from adhering after operation.
