摘要
目的观察芦丁(Rutin)对人肝癌HepG2的生长和增殖的影响。方法体外培养HepG2细胞,用甲基塞唑基四唑(MTT)法、3^H—TdR掺人法测定芦丁对人肝癌HepG2的生长和增殖抑制情况,并且用荧光染色法在Olympus荧光显微镜下观察芦丁对HepG2细胞凋亡的影响,流式细胞仪检测细胞周期变化。结果芦丁能抑制HepG2细胞的生长、增殖、诱发细胞凋亡;在50~250μmol/L浓度范围内处理的HepG2,G0/G1期细胞增加,G2/M期细胞减少,细胞周期阻滞于G0/G1期。结论芦丁能抑制HepG2细胞的生长,并诱发细胞凋亡,呈浓度依赖性。
Objective To observe the effect of rutin on growth and proliferation of human hepatic cancer line (HepG2). Methods HepG2 cells were cultured in vitro, then cocuhured with 50 to 250 μmol/L rutin for 24 h. The inhibition rate of rutin on growth and proliferation of HepG2 was determined by MTT, 3^ H-TdR, and apoptotic cells were observed in fluorescent staining by Olympus fluorescent microscopy, and cell cycle was analysed by flow cytometry. Results Rutin inhibited HepG2 cells from growth and proliferation, and evoked apoptosis. Flow cytometry showed that 50 to 250 p, mol/L rutin caused an increase at G0/G1 phase and a decrease at G2/M phase and arrest at G0/G1 phase in the cell cycle. Conclusion Rutin markedly inhibits the proliferation of HepG2 cells and induces apoptosis in a concentration-dependent manner.
