SSTF预处理对缺血再灌注心肌超微结构及细胞凋亡的保护作用

Protective Effect of Scutellaria Baicalensis Stem-leaf Total Flavonoid Against the Myocardial Ischemia Reperfusion on Myocardial Cell Ultrastructure in Rats

摘要 目的:探讨黄芩茎叶总黄酮(SSTF)预处理对缺血再灌注大鼠心肌超微结构的影响及其作用机制。方法:40只Wistar大鼠分为SSTF预处理组(SSTF组)、缺血再灌注组(IR组)和假手术组。SSTF组于术前1周灌服SSTF(50mg、100mg、200mg.kg-1.d-1),另2组给等量PBS。制备IR模型,光、电镜观察各组大鼠心肌超微结构变化和心肌细胞凋亡指数(AI)。结果:SSTF各组均较IR组心肌细胞超微结构尤其是线粒体损伤程度减轻,且呈一定剂量依赖效应,心肌凋亡细胞减少。结论:SSTF预处理对缺血再灌注的心肌细胞具有保护作用,其机制可能是通过SSTF稳定心肌细胞膜和线粒体,ATP的生成增多,拮抗Ca2+和清除氧自由基来实现的。 abjctive: to explore the affect and effective mechanism about SSTF pretreatment against the myocardial ischemia reperfusion on myocardial ultrastructures. Methods: The male 40 wistar rats were divided into SSTF membranaceus pretreatment group, ischemia reperfusion group and false operation control group. Rats of SSTF group were perfused with SSTF, others two groups were given PBS solution instead of SSTF equally at the a week before the operation. The ischemia reperfusion modal were prepared and the specimen were made into slices from the model, The ultrastructural alterations and the index of cell apoptosis were observed under the transmission electron microscopy. Result: The degree of injuring on uhrastructures of myocardial cell in SSTF group is rather alleviated than in ischemia reperfusion group and presented a dosedependence effective relations, the myocardial cell apoptosis is decreased. Conclusion: it is proved that the SSTF pretreatment can protect the myocardial cell against the myocardial ischemia, the protective mechanism may be obtained by stabilizing the cell membrane and the mitochondria and by regenerating more ATP, againsting the Ca^2+, and by getting rid of oxygen free radical.
