人工血管内皮细胞衬附细胞基质物的筛选 被引量:2

摘要 采用析因设计及LUZEX-F型彩色图像分析仪判定内皮细胞融合度等手段,研究明胶,纤维连接蛋白(fibronectin,FN),人血浆和20%人体白蛋白,单一或混合物,共31种做为预衬物,对促进人脐带静脉内皮细胞贴附Dacron人工血管生长的作用。结果:1%明胶+血浆+300μg/ml纤维连接蛋白、1%明胶+血浆+750μg/ml纤维连接蛋白、1.5%明胶+750μg/ml纤维连接蛋白三种预衬物对促进人脐带静脉内皮细胞贴附Dacron人工血管生长的功效最好。同时反映出:随明胶浓度的升高,促进内皮细胞贴壁生长的能力增强;明胶与血浆、明胶与FN、明胶与20%人体白蛋白及血浆与20%人体白蛋白间有的较强的协同促进内皮细胞贴壁生长的作用;FN与20%人体白蛋白混合不但没有协同促进内皮细胞贴壁生长的作用,反而会影响其它因素间协同促进作用;20%人体白蛋白单独做为预衬物没有促进内皮细胞贴壁生长的作用。 Division factor analysis design and LUZEX F type color image analysis computer in measuring the confluence of endothelial cells(EC) were used to investigate the effect of thirty one kinds of precoating substances, which come from monoconcentrations or combinations of gelatin (0.5%, 1%, 1.5%), fibronectin (300 μg/ml,750 μg/ml), human plasm and 20% human albumin, on improving the attachment and proliferation of human umbilical vein endothelial cells (HUVEC) on Dacron artificial vascular surface. The results indicated that 1% gelatin+human plasma +300 μg/ml fibronectin, 1% gelatin+ human plasma+750 μg/ml fibronectin, were the best of all the precoating substances, which have better effect on improving the attachment and porliferation of HUVEC. The results also showed that the gelatin's effect on the attachment and proliferation of HUVEC got better when its concentration increased. Gelatin and plasma, gelatin and fibronectin, gelatin and 20% human albumin, plasma and 20% human albumin had strong synergistic effect on improving EC attachment and proliferation whereas fibronectin and 20% human albumin not only had not the synergism on improving the attachment and proliferation of EC, but decrease the effect of other procoating substances. Twenty percent of human albumin alone had no effect on improving the attachment and proliferation of EC.
