激动素对肝纤维化大鼠肝脏胶原变化的作用研究

摘要 目的:观察激动素(kinetin)对肝纤维化大鼠肝脏Ⅰ、Ⅲ型胶原含量的作用。方法:将实验大鼠随机分为3组,模型组用CCl4诱导形成肝纤维化模型;激动素组造模同时给予0.1%激动素溶液5ml·kg-1·d-1;对照组仅给予生理盐水皮下注射,均治疗12周。应用苦味酸天狼猩红偏振光法和图像分析技术对3组中的胶原含量及分布特点进行观察。结果:激动素组Ⅰ型胶原与Ⅲ型胶原含量分别为(2.107±0.833)%和(1.114±0.413)%,分别较模型组(4.473±2.311)%和(1.964±0.854)%显著降低(P<0.01)。结论:激动素能使肝纤维化大鼠肝脏Ⅰ、Ⅲ型胶原含量显著降低。 Objective: To detect the effect of kinetin on type I and III collagen fibrils in rats with hepatic fibrosis. Methods: Rats were randomly divided into 3 groups: model group with CCIA induced hepatic fibrosis; Kinetin group were given 0. 1 % kinetin 0. 5 ml/100 g per day at the time of making model; control group were given normal saline subcutaneously. All the groups were treated for 12 weeks. The content and distribution of collagen fibrils were observed with both picric-sirius red polarimetry and image analysis. Results: The content of type Ⅰ and Ⅲ collagen fibrils were (2. 107 ± 0. 833) % and ( 1.114 ± 0. 413)% respectively in the kinetin group, significantly lower than those in the model group (4. 473 ± 2. 311) % and (1. 964 ± 0. 854) % respectively (P〈0. 01). Conclusion: Kinetin may significantly decrease the content of type Ⅰ and Ⅲ collagen fibrils in rats with hepatic fibrosis.
