摘要
目的进一步探讨人乳腺癌组织中微钙化的形成机制。方法收集乳腺癌组织30例,同时取距癌组织边缘5cm以上非癌正常乳腺组织作对照。患者术前均行钼靶X线检查且临床病理资料齐全。应用半定量RT-PCR方法检测并比较了组织中OPN和ONmRNA的表达水平,分析了其与微钙化表现和各临床病理特征的关系。结果乳腺癌组织与正常乳腺组织中OPNmRNA校正值(OPN/β-actin)分别为0.479±0.114和0.205±0.059;ONmRNA校正值(ON/β-actin)分别为0.480±0.109和0.223±0.049。两组比较差异显著(P<0.05)。本组病例钼靶片中表现微钙化13例,占43.30%,钙化病例电泳条带亮度明显高于无钙化者,扩增结果显示钙化组与无钙化组OPNmRNA校正值分别为0.604±0.036和0.385±0.022;ONmRNA校正值分别为0.598±0.036和0.389±0.020。两组比较有显著性差异(P<0.05)。钙化组与正常乳腺组相比,差异更显著(P<0.05)。乳腺癌组织中OPN、ONmRNA表达与患者年龄、腋下淋巴结有无转移、肿瘤分化程度和临床分期均无显著相关性。结论乳腺癌组织中OPN和ONmRNA表达水平与微钙化的出现密切相关,可能参与和/或调节了微钙化的形成。
Objective To further explore the mechanism of the formation of microcalcification in breast cancer tissues. Methods Fresh specimens of cancer tissue and normal breast tissue adjacent to cancer (〉5 cm) from thirty cases of breast cancer were obtained. All the patients were with complete mammography and clinical data. The expression of OPN and ON mRNA was semi-quantitively detected by RT-PCR. The relationships with microcalcification and clinicopathologic features were analyzed. Results The ratio of OPN and ON to β-action in breast cancer and normal breast tissues was 0. 479±0. 114, 0. 205±0. 059; 0480±0. 109, 0. 2335±0. 049 respectively. The difference in ratios of both OPN and ON toβ-action between cancer and normal tissues was significant (P〈0.05). the brightness of electrophoretic bands of the 13 cases with microcalcification on mammogram was obvious higher than that in the cases without microcalcification. The ratio of OPN and ON to β-action of microcalcification group and without microcalcification group was 0. 604±0. 036, 0. 385 ± 0. 022; 0. 598 ± 0. 036, 0. 389±0. 020 respectively, there was a significant deviation (P〈0.05). Compared with normal breast tissues, the difference was more obvious (P〈0.05). There was no obvious difference between the expression of OPN and ON mRNA and clinical pathological features, such as age, clinical staging, pathological grade and status of axillary nodes metastases. Conclusion The expression of OPN and ON mRNA of breast cancer tissue was well correlated to the microcalcification, they maybe participate in and/or adjust the form of microcalcification.
