摘要
目的建立高效液相色谱法测定盐酸多西环素脂质体凝胶中盐酸多西环素的含量。方法采用RP-HPLC法,色谱柱为十八烷基硅烷键合硅胶柱,以0.05mol·L^-1草酸铵溶液-二甲基甲酰胺-0.2mol·L^-1磷酸氢二铵溶液(65:50:5),用氨试液调节pH8.0±0.2为流动相;柱温40℃;在280nm波长处检测。结果多西环素在0.3298~16.4920μg·mL^-1浓度与峰面积呈良好的线性关系(r=0.9998),方法平均回收率为99.9%(n=3),RSD为1.18%。结论该法简便、可靠、专属性强,适合于该制剂的含量测定及产品的质量控制。
OBJECTIVE To establish an HPLC method for the determination of doxycycline hyclate in the gel of doxycycline hydrochloride liposome. METHODS A C18 column was used. The mobile phase consisted of 0. 05 mol·L^-1 ammonium oxalatehexane and DMF and ammonium phosphate dibasicthe (65 : 50 : 5), and the detection wavelength was 254 nm. RESULTS The linear range was 0. 3298~16. 492μg·mL^-1 (r=0. 999 8). The average recovery of the method was 99.9%, and RSD was 1.18% (n= 3). CONCLUSIONS This method is simple, convenient and suitable for the determination and the quality control of doxycycline hyclate in the gel of doxycycline hydrochloride liposome.
