摘要
目的建立高效液相色谱法测定环孢霉素眼用原位凝胶的含量。方法色谱柱为Thermoelectory C18,流动相:乙腈-水(90:10),检测波长210m,流速0.7mL·min^-1。样品用甲醇溶解并用0.45μm微孔滤膜过滤。结果在5~60μg·mL^-1范围内,环孢素对照品的峰面积(Y)与其浓度(X)呈良好的线性关系,r=0.9996,高、中、低3种浓度平均回收率99.3%、99.8%、98.9%(n=3),日内、日间RSD分别为1.45%,1.93%(n=5)。结论该法快速、灵敏、准确、专属性高。
OBJECTIVE To determine the contents of cyclosporine in ophthalmic situ-forming gel with HPLC. METHODS A reversed phase high performance liquid chromatographic method was developed. Cyclosporine was chromatographed on thermoelectory C18 column and detected at 210 nm. The mobile phase was acetonitrile., water (90 : 10), the flow rate was 0.7 mL · min^-1. Samples were dissolved with methanol, and then was filtered with 0.45μm filter membrane. RESULTS The linearity of cyclosporine was 5~60μg·mL^-1 and the coefficient of correlation was 0. 999 6. The average recoveries of cyclosporine were 99.3%, 99.8%, and 98.9%(n=3). The within day and between day RSD were 1.45% and 1.93% (n=5) respectively. CONCLUSIONS The method is rapid, sensitive and efficient for the determination of CsA in ophthalmic situ-forming gel.
