摘要
目的 研究重组巴曲酶(rBAT)对动物凝血系统的作用。方法 通过测定实验动物的出血时间(BT)、凝血时间(CT)和血栓干、湿重,观察rBAT对正常小鼠BT,CT的影响和对正常小鼠止血作用的时效关系;rBAT对肝素和尿激酶所致的小鼠出血倾向模型的治疗作用;以及rBAT对正常大鼠体内结扎静脉血栓形成的影响。结果rBAT0.5—3.3KU·kg^-1可使正常小鼠BT,CT显著缩短,但剂量加大至10.0Ku·kg^-1时又使BT,CT延长;其对正常小鼠的止血作用可持续2h以上;rBAT对于肝素和尿激酶所致的小鼠出血倾向具有一定的逆转作用;rBAT3.0—10.0KU·kg^-1可促进大鼠体内血栓形成,30.0KU·kg^-1则起抑制作用。结论 rBAT对实验动物的凝血系统具有双向作用。
Aim To study the effect of the recombinant batroxobin (rBAT) on blood coagulation system. Methods The effect of rBAT on bleeding time (BT) and blood coagulation time (CT) in normai mice and hemorrhage mice modeled by heparin or urokinase were observed respectively. The time-effect relationship in mice was investigated. The weight of thrombus that formed in ligated inferior caval vein in rats was also3 measured. Results rBAT significantely shortened BT and CT in mice at the dose of 0.5 -3.3 KU·kg^-1 When the dose increased to 10 KU·kg^-1 BT and CT were prolonged, rBAT could reverse the hemorrhagic tendency induced by heparin or urokinase to some degree, rBAT could promote the thrombosis in ligated inferior caval vein in rats, At the dose of 3.0 - 10.0 KU·kg^-1 ,inhibit the thrombosis and at the dose of 30.0 KU·kg^-1. Conclusion rBAT has two-side effect on blood coagulation system.
