摘要
目的建立以反相高效液相色谱法(RP-HPLC法)测定雷米普利血药浓度的方法。方法用固相萃取小柱提取雷米普利血浆样品,以水杨酸为内标,流动相为乙腈-pH值为2.0的磷酸盐缓冲液(30∶70),柱温为60℃,检测波长为210nm,流速为1.0mL/min。结果雷米普利浓度在5~100ng/mL范围内与峰面积线性关系良好,日内和日间RSD<15%(n=5)。结论RP-HPLC法具有较高的准确性、精密性和灵敏度,且操作简便、快速,适用于雷米普利血药浓度监测。
Objective To establish a RP- HPLC method for the determination of ramipril in human plasma. Methods Using salicylic acid as the internal standard,ramipril was extracted from the human plasma with solid-phase extraction cartridge. The mobile phase was acetonitrile- pH=2.0 phosphonic acid buffer salt (30:70),the column temperature was 60℃ ,the detecting wavelength was 210 nm and the flow rate was 1.0 mL/min. Results The calibrating curves of ramipril were linear in the range of 5- 100 ng/mL. The within- day and between- day RSD were less than 15% (n = 5). Conclusion The method is accurate,sensitive and convenient for ramipril monitoring in human plasma.
