摘要 采用1,6-二苯基-1,3,5-己三乙烯(DPH)荧光偏振法研究了镉在小鼠体内对肝线粒体膜流动性的影响,同时观察了膜结合酶活性和线粒体呼吸功能的变化。结果表明,镉能显著增加小鼠肝线粒体膜的荧光偏振度(P)及微粘度(η),并导致线粒体呼吸控制比(RCR)和氧化磷酸化效率(ADP/O)显著下降。膜结合K+、Na+-ATP酶、Ca2+-ATP酶、Mg2+-ATP酶和琥珀酸脱氢酶(SDH)的活性均受到明显抑制。 The effect of cadmium on the membrane fluidity of hepatic mitochondria in mice were investigated by spectrofluorometer using DPH as a fluorescence probe. Membrane bound enzyme activity and respiratory function of mitochondria were also measured. It was shown that cadmium prominently increased the fluorescence polarization (P) and microviscosity (η) of mitochondrial membrane and significantly decreased RCR and ADP/O of mitochondria with the activity of membrane bound ATPase and SDH being significantly inhibited. The results suggested that selenium might antagonize aforementioned changes induced by cadmium.
