摘要
目的 评估TIGER细胞图像分析仪测量细胞图像光度的线性.方法 选取十只健康成年小白鼠的睾丸组织,制成细胞悬液,涂片,快速Feulgen染色,TIGER细胞图像分析仪测量涂片内三种单个完整生精细胞核的DNA含量并比较它们的比值关系,分析其平均吸光度(AOD)的最大、最小值范围.结果 三种生精细胞核的DNA含量呈1、2、4的比例,其AOD值范围为0.73~1.64.结论 TIGER细胞图像分析仪在其平均吸光度值为0.73~1.64的范围内测得的组织、细胞图像光度呈线性.
Objective To evaluate the linearity of photometry for measuring cell image of TIGER cell image analysis system. Methods Smears of spermatogenic cell suspension from ten healthy mice were stained with quick Feulgen stain. DNA contents of single intact nucleus of the three sorts of testicular spermatogenic cells on the smear were measured with TIGER cell image analysis system. Then the ratio of the nuclear DNA contents among spermatid, spermatogonium and spermiocyte were compared and their extension of AOD was analyzed. Results The ratio of DNA content among spermatid, spermatogonium and spermiocyte was close to 1, 2, 4. Their extension of AOD was 0.73 - 1.64. Conclusion TIGER cell image analysis system satisfies with linear requirements of photometry of tissue and cell images. In the extension of AOD 0.47 - 1.64, photometry of tissue and cell images is linear.
