甲基丙烯酸环氧丙酯对BALB/c 3T3细胞恶性转化的实验研究 被引量:10

摘要 利用建立的 BALB/c 3T3细胞转化系统进行甲基丙烯酸环氧丙酯(GMA)的细胞转化和裸鼠诱癌的实验研究。实验结果表明:GMA 可诱导 BALB/c 3T3细胞形态转化,并呈剂量-反应关系;从转化灶分离扩增细胞,经电镜扫描观察到转化细胞表面形态改变,可被植物凝集素 ConA凝集,并在半固体琼脂生长,在同品系裸鼠皮下形成了肿瘤,病理组织检查结果为分化程度较低的纤维肉瘤。GMA 有很强的诱导细胞恶性转化的能力,提示 GMA 有潜在的致癌危险性。 BALB/c 3T3 cells were used for the study on malignant transformation induced by glycidyl methacry- late (GMA).The results showed that the response of the cell to GMA was in a dose—response dependent manner.A morphological transformation of BALB/c 3T3 cells were induced by GMA.The changes on the surface of transformed ceils were observed under screening electroscope.The transformed cells were agglu- tinated following exposure to ConA and the colony formed on semisolid agar medium.Fibrosarcoma were grown in the homologous nude mice after injection with the transformed cells.The malignant transformation of BALB/c 3T3 cells induced by GMA will suggest the potential carcinogenesis of this compound.
