摘要 目的复制经典的酒精性肝损伤动物模型,为防治酒精性肝损伤的实验研究提供理想的研究工具。方法根据Lieber-DeCarli酒精液体饲料配方配制含酒精和不含酒精而热卡相同的液体饲料。23只大鼠随机分正常对照组(n=5)、无酒精液体饲料组(n=9)、酒精液体饲料组(n=9)。正常对照组大鼠给予常规饲料,无酒精液体饲料组与酒精液体饲料组分别定量给予相应液体饲料自动饮服。8周后,观察大鼠肝组织病理形态、肝细胞超微结构的变化,血清丙氨酸氨基转移酶(alanineaminotransferase,ALT)、天冬氨酸氨基转移酶(aspartateaminotransferase,AST)以及肝组织γ-谷氨酰转肽酶(γ-glutamyltransferase,γ-GT)活性、甘油三酯(triglyceride,TG)含量,以及脂质过氧化损伤相关指标的变化。结果酒精液体饲料组大鼠血清ALT、AST活性,肝组织TG含量以及脂质过氧化产物较正常对照组和无酒精液体饲料组有较大幅度的升高,同时抗氧化物质减少;酒精液体饲料组肝细胞肿胀、脂肪变性等病理改变严重。结论自行配制Lieber-DeCarli液体饲料可成功复制出早期酒精性肝损伤病理改变的大鼠模型。 Objective: Duplicating the classical alcoholic hepatic injury model, to provide an ideal animal model for research on prevention and treatment of hepatic injury. Methods: According to the prescription of Lieber-DeCarli, the same calorie fluid animal feed, which contained ethanol or non-ethanol, was prepared. Twenty-three rats were divided into normal control group (n = 5), control liquid diet group (n=9), ethanol liquid diet group (n = 9). Rats in the normal control group were fed normal diet, and rats in the control liquid diet group and ethanol liquid diet group were fed the corresponding diet for eight weeks. The pathologic changes of hepatic tissue were observed. The activities of the serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), the liver tissue γ-glutamyltransferase (γ-GT), the content of triglyceride (TG), and the lipid peroxidation by-products were assayed. Results: Compared to the normal control and the control liquid diet groups, the activities of ALT, AST, and γ-GT, and the lipid peroxidation by-products increased significantly in the ethanol liquid diet group. The pathological changes of cellular swelling and fatty degeneration in the ethanol liquid diet group were severe. Conclusion: Alcoholic hepatic injury model can be successfully duplicated by Lieber-DeCarli prescription.
