摘要
目的:研究冰片对培养的大鼠海马神经元GABAA受体介导电流的调控。方法:采用全细胞膜片钳方法。结果:在培养的海马锥体神经元上,给予GABA可引起一具有明显脱敏的内向电流(IGABA),此电流能被b icucu llin所阻断,说明该电流为GABAa受体所介导。冰片以open-channel b locker的方式抑制IGABA,且这种抑制作用是可逆的,即洗去冰片后,抑制作用迅速消失。结论:冰片可能通过抑制GABAA受体提高中枢神经系统的兴奋性而发挥其醒脑作用。
Objective: To study the modulatory effect of Bomeolum Syntheticum on GABA-activated current(IGABA) in cultured rat hippocampal neurons. Methods: Using the whole-cell patch-clamp technique. Results: Application of GABA evoked inward currents which had an obvious desensitization. These GABA-activated currents were reversibly blocked by the GABAa receptor antagonist bicuculline. These results indicated that the response to GABA were mediated by activation of GABAA receptors. Borneolum Syntheticum can inhibit IGABA reversibly acts as an open-channel blocker. Conclusion: These results suggest that Borneolum Syntheticum could up-modulate the CNS excitability by inhibition of the GABAA receptor-mediated current.
