摘要
目的探索一种可弥补经典双足鼠模型伦理学缺陷的改良手术方法。方法 20只 SD大鼠随机均分为改良组和传统组,改良组行臂丛神经离断,传统组行近肩关节截肢。改良手术前后分别观察肩关节主动运动并记录肩关节周围肌肉肌电图以确定肌肉麻痹范围。对比改良法与传统建模法的手术持续时间、腋动脉损伤概率评价手术风险。记录术后2 d、1、2、4、6周双足鼠日均累计站立时间及6周站立姿势的差异,评价改良术式所构建双足鼠动物模型模拟人类站立姿势的效果。结果改良法较传统截肢手术操作简化,手术时间缩短,腋血管损伤概率显著降低。改良法术后肌电图显示所有上肢肌及大部分肩关节周围肌肉去神经化,改良双足鼠肩关节仅遗留轻微外展运动功能,上肢完全瘫痪。手术2周后,改良双足鼠日均累计站立时间甚至长于经典双足鼠,而直立姿势与经典双足鼠无差异。结论双足鼠改良建模手术方法不但简化了手术操作,而且实现了较经典截肢术更佳的建模效果,同时彻底驱除了双足鼠模型的伦理学阴霾。
Objective To explore an improved operation approach to make up the ethical deficiency of classical bipedal rat model. Methods 20 SD rats were randomly divided into an improved group and atraditional group averagely. Improved group were operated with plexus brachialis nerve amputation, while traditional group with forelimbs amputation close to shoulder joint. Forelimb and shoulder joint movements were observed and electromyographs of the peripheral muscles around shoulder joint were recorded to make certain the paralytic scope. Operation duration and axillary artery hurt probability of both the improved and traditional operation were compared to evaluate the operation risk of improved approach. Mean total upright time on 2 d, 1 w, 2 w, 4 w and 6 w post-operation and upright posture on 6w post-operation were recorded to evaluate the human-upright-posture-simulating effect of bipedal rats operated with improved approach. Results Compared with classical amputation approach, improved approach exhibited simpler operation procedure, shorter operation duration and lower axillary artery hurt probability. Electromyographs revealed that all forelimb muscles and most shoulder-peripheral muscles were denervated, forelimbs of the improved bipedal rats were absolutely paralyzed and shoulder joint reserved only slight abduction function. The mean total upright time of improved bipedal rats was longer than the classical ones after 2 w post-operation, while upright posture showed no difference with classical bipedal rats. Conclusions The improved operation approach not only simplified operation procedures, but also achieved even better effect than classical amputation operation and exempted bipedal rat model from the ethical shadow.
