水通道蛋白-4对血吸虫病性脑血管通透性的作用 被引量:1

摘要 目的:观察水通道蛋白-4(AQP-4)对脑血吸虫病脑血管通透性的作用。方法:采用尾蚴腹贴法复制兔血吸虫病模型。45天后杀虫并随机分为手术组、假手术组、对照组(n均=14)。采用颈内动脉虫卵注射法复制脑血吸虫病模型。术后7天取脑组织。光镜、电镜观察形态学改变;免疫组织化学观察AQP-4在脑中的表达变化。结果:免疫组化显示,AQP-4主要表达在脉络丛、室管膜、软脑膜及血管周围的星形胶质细胞。手术组AQP-4的表达量与假手术组及对照组的差异有统计学意义(P<0.05)。对应的形态学改变为持续的脑组织血管源性水肿。结论:脑血吸虫病脑水肿中,AQP-4表达升高;降低血管的通透性,可能起潜在的保护作用。 Objective: To investigate function of aquaporin-4(AQP-4) on vascular permeability in encephalopathy of schistosomiasis.Method: 42 rabbits were infected percutaneously by cercarie of Schistosoma japonicum.45 days later,they were administrated schistosomicide,then randomly divided into operation group,fake-operation group and control group averagely(n=14).Schistosoma eggs were injected in internal carotid artery to construct the model of encephalopathy of schistosomiasis in operation group.Tissue of brain was obtained after 7 days.Optic microscopy and transmission electron microscopy were applied to detect the pathologic features of brain tissue.The expression and distribution of AQP-4 in the brain were analyzed by immunohistochemistry.Results: The AQP-4-positive parts mainly included choroid plexus,ependyma,pia-mater cranialis,perivascular astrocytes.There was a significant higher expression of AQP-4 in operation group than that in other groups.The corresponding pathologic morphology alteration was persistent vasogenic edema.Conclusion: It demonstrated a potential protective effect of AQP-4 on encephalopathy of schistosomiasis,suggesting that AQP-4 induction may reduce vascular permeability.
