摘要
目的采用反相高效液相色谱(RP-HPLC)法测定车前草中主要成分乌索酸和齐墩果酸的含量。方法采用高效液相色谱法。使用Nova-Pak C18柱(3.9mm×300mm,4μm),流动相为甲醇:水(88:12,V/V),流速为0.8ml/min,检测波长为210nm,柱温25℃。结果乌索酸在0.456~4.104μg范围内呈良好的线性关系(r=0.9997),平均回收率为98.2%,RSD=1.1%;齐墩果酸在0.156~1.384μg范围内呈良好的线性关系(r=0.9996),平均回收率为101.6%,RSD=1.7%。结论方法灵敏、准确、专属性强,重现性好,可作为车前草药材的质量控制方法。
Objective To establish a method for determining ursolic acid and oleanolic acid in Plantago asiatica L. by high performance liquid chromatography. Methods The separation was carried out on Nova- Pak C18 column (3.9 mm× 300 mm, 4 μm) with methanol -water (88:12,V/V) as the mobile phase,at a flow rate of 0.8 ml/min. The detection wavelength was set at 210 nm. The column was used at 25 ℃. Results Ursolic acid showed a good linear relationship in the range of 0.456 -4. 104 μg (r = 0.9997) and the average recovery was 98.2 %, RSD was 1.1% . Oleanolic acid showed a good linear relationship in the range of 0. 156 -1.384 μg (r =0.999 6) and the average recovery was 101.6 %, RSD was 1.7%. Conclusion The method is sensitive,accurate and specific with a good repeatability and can be used for the quality control of Plantago asiatica L.
