摘要
目的:探讨内皮素(ET)与肾小球肾炎的关系,方法:建立改良慢性血清病大鼠系膜增生性肾炎动物模型,采用 ̄125I-ET放射免疫分析法测定肾炎组与付照组右心血和肾静脉血中FT-1的水平。应用免疫组化及免疫电镜方法进行FT-1的定位研究,结果:肾炎组肾静脉血及右心血FT-1血浆水平均高于对照组(P<0.05),而且肾静脉血FT-1含量明显高于右心血(P<0.001)。FT-1广泛存在于肾脏的各种固有成分中。结论:肾炎时肾阻自身合成和分泌的FT是血浆FT的主要来源,炎性增生的肾小球固有细胞和肾小管上皮细胞分泌的FT是肾脏局部FT的主要来源。
Objective:To explore the ralationship between endothelin (ET)and glomerulonephritis.Methods:Rat model of mesangioproliferative glomerulonephritis(MsPGN)was established.The ET-1 levels in the right ventricle plasma were measured and compared with those in renal vein by125I-radioim-munoassay(RIA).The distribution of FT-1 in kidney was detected by immunohistochemical staining and immunoelectron-microscopy.Results:The plasma level of FT-1 was significantly greater in experi-mental group than that of control group both in the ventricle and renal vein.Level of ET-1 in renal vein was significantly higher than that in the right ventricle,Immunohistochemistry staining and immuno-electron-microscopy showed that ET-1 widely disposed many components of kidney. Conclusion:The plasma of ET-1 derivey mainly from proliferative mesangial cells and renal tubular secretion in MsPGN.
