TGF-β_1在db/bd自发性糖尿病小鼠颌下腺表达的研究

摘要 目的探讨db/db自发性糖尿病小鼠颌下腺转化生长因子β1(transforming growth factor-β1,TGF-β1)表达与其形态学改变的关系。方法引进日本表型正常隐性基因小鼠,近亲交配,其纯合子后代,即为db/db(单基因遗传自然发病型)型糖尿病小鼠。取3、4、6、8、10月龄db/db糖尿病小鼠颌下腺(实验组)及相应月龄的db/+m正常小鼠颌下腺(对照组)。每组五只,HE及SP免疫组化染色后行图像分析,光镜观察颌下腺的病理形态学改变,统计TGF-β1阳性表达的细胞数。结果随着糖尿病发展,颌下腺实质组织萎缩,细胞缩小,形态不规则,排列不整齐,间质纤维增生,TGF-β1阳性细胞数逐渐增加,呈上升趋势,且明显高于同龄对照组p<0.01。结论db/db糖尿病使小鼠颌下腺TGF-β1表达增强,同时有间质纤维增强,TGF-β1表达上调可能与糖尿病间质纤维增生密切相关。 Objective To investigate the relationship between morphological change and expression of transforming growth factor- β1 in submandibular gland from C57BK/ksj - db/db mutant diabetic mice. Methods Animals were C57BL/ksj - db/+ m mutant diabetic mouse, which were imported from Japan. The pure zygote, it is db/db mouse, displays obesity and high blood glucose over 10mmol/L. The experimental group selected 3,4,6,8,10 month- old db/db mouse and the age - matched db/+ m mouse as the control, lmmunohistoehemistry technology was used in this study. Results In db/db diabetic mice, submandibular gland had atrophied and showed the significant proliferation of interstitial fibers. The number of TGF-β1 immunopositive cells in submandibular gland was significantly increased than that of age- matched controls and increased with the diabetes progression. Conclusion db/db matant diabetes made the results of expression of TGF -β1 increased and interstitial fibers proliferated in submandibular gland. The increased TGF - β1 expression may be associated with proliferating intersitial fiber.
