orf224基因与芸薹属种间杂种雄性不育性的相关性 被引量:5

摘要 orf224是在甘蓝型油菜(Brassica napus)波里马(Polima)胞质中发现的一个与细胞质雄性不育(Cytoplasmicmale sterility,CMS)相关的线粒体基因.大白菜(B.campestrisssp.pekinensis)CMS系CMS3411-7是由Polima不育系和大白菜自交系3411-7杂交后,以3411-7为轮回亲本经多代回交选育得到的.以CMS3411-7为母本分别与小白菜(B.campestrisssp.chinensis)自交系S71、大白菜自交系K1和菜心(B.campestrisssp.chinensisvar.utilis)自交系B1杂交得到3个杂交种CS71、CK1和CB1.以CMS3411-7、CS71、CK1和CB1为材料,以Polima为对照,利用多聚酶链式反应(Polymer-ase chain reaction,PCR)技术与田间实验育性观察相结合的方法,研究了orf224与几个芸薹属种间杂种雄性不育性之间的相关性.结果显示:在这4个材料中尽管均含有orf224基因,但CMS3411-7和CB1表现为雄性不育,CS71和CK1表现为雄性可育.这就表明,orf224虽是CMS的因子,但它必须和核基因进行互作才能发挥作用. orf224 is a mitochondrial gene discovered in Polima cytoplasm of rapeseed (Brassica napus) ,which is considered to be associated with cytoplasmic male sterility (CMS). CMS line CMS3411-7 of Chinese cabbage(B, campestris ssp. pekinensis)was produced by cross pollination and repeated backcrosses between Polima and Chinese cabbage inbred lines 3411-7 as recurrent male parent. Using CMS3411-7 as female parent, three F1 hybrids CS71 ,CK1 and CB1 were derived from the cross of CMS3411-7 with Pakchoi (B. carnpestris ssp. chinensis)inbred line Sn ,Chinese cabbage inbred line K, and flowering Chinese cabbage (B. carnpestris ssp. chinensis var. utilis) inbred line B1, respectively. In this paper, using CMS3411-7, CS71, CK1 and CB1 as materials,Polima as control, the correlation of the orf224 gene with cytoplasmic male sterility of several interspecific and inter-subspecific hybrids in Brassica was studied by means of PCR and pollen fertility observation. The results showed that all the four materials contained orf224 gene,but CMS3411-7 and CB1 were male sterile,CSn and CK1 were male fertile. These results indicated that orf224 was one of the factors of CMS,but orf224 must interact with nuclear gene to produce function.
