艾溃灵合剂促进模型豚鼠口腔溃疡恢复的效果评估 被引量:10

摘要 目的:观察具有清热泻火,解毒化瘀,益气养阴功效的艾溃灵合剂治疗口腔溃疡的特点。方法:实验于2005-02/10在河南中医学院动物实验中心完成。以苯酚灼烧豚鼠面颊造口腔溃疡模型为实验对象,将动物按照随机数字表法分为6组,即艾溃灵合剂20,10,5g/kg剂量组(黄芪、生地、白花蛇舌草、连翘、甘草等药物组成),口炎清颗粒剂组(33.3g/kg,天冬、麦冬、玄参、金银花、甘草等药物组成),模型组及空白组,每组10只。除空白对照组外,其余5组造口腔溃疡模型,每日涂抹及灌胃给药,涂艾溃灵合剂各剂量组给药浓度分别为1,0.5,0.25g/mL,涂量为0.5mL,灌服20mL/kg;口炎清颗粒剂组给药浓度为0.167g/mL,涂量为0.5mL,灌服10mL/kg;模型组涂抹生理盐水0.5mL,灌服生理盐水10mL/kg;空白组不予处理。每天给药1次,连续用药5天,于第1,3,5天用数码照像头摄下豚鼠口腔溃疡情况,于最后1次用药后2h,取下口腔溃疡局部,作病理切片,观察溃疡局部充血水肿程度以及溃疡模型组织形态。结果:豚鼠60只均进入结果分析。①用药第3天,艾溃灵合剂20,10,5g/kg剂量组,口炎清颗粒组与模型组比较,均可使溃疡面积显著减少[(11.8±3.2),(8.9±6.6),(13.4±4.7),(13.0±5.8),(20.30±4.6)mm2;P<0.01]。第5天,艾溃灵合剂20g/kg剂量组溃疡面积与模型组比较,差异有非常显著性意义[(3.9±5.6),(12.2±6.2)mm2;P<0.01],艾溃灵合剂10,5g/kg剂量组,口炎清颗粒组溃疡面积与模型组比较,差异有显著性意义[(6.1±3.0),(7.0±3.6),(6.3±3.3)mm2;P<0.05]。②用药第3天,艾溃灵合剂20,10g/kg剂量组和口炎清颗粒组均可显著减轻口腔溃疡程度(P<0.01);用药第5天,20g/kg剂量艾溃灵合剂组可显著减轻口腔溃疡程度(P<0.01),艾溃灵合剂10g/kg剂量组和口炎清组可明显减轻口腔溃疡程度(P<0.05),艾溃灵合剂20g/kg剂量组口腔溃疡程度较口炎清组减轻(P<0.05)。③艾溃灵合剂20,10g/kg剂量组和口炎清颗粒组口腔溃疡组织病理变化显著减轻(P<0.01),艾溃灵合剂5g/kg剂量组仅有减轻口腔溃疡病理变化的趋势。结论:艾溃灵合剂可显著减轻豚鼠口腔溃疡症状,明显促进豚鼠口腔溃疡的恢复。20g/kg艾溃灵合剂效果优于口炎清颗粒。 AIM: To observe the characteristics of Aikuiling liquid, which can clear heat and purge fire, remove the toxic substances and blood stasis, and supplement qi and nourish yin, in treating oral ulceration. METHODS: The experiment was conducted in the animal experiment center of Henan College of Traditional Chinese Medicine from February to October 2005. The guinea pigs, whose cheeks were burnt by phenol to establish the models of oral ulceration, were selected as subjected and randomly divided into 6 groups: 20, 10, 5 g/kg doses of Aikuiling (composed of milkvetch root, rehmannia dride rhizome, Hedyotic diffusa, forsythia suspense and licorice root) groups, Kouyanqing granule group (33.3 g/kg, composed of asparagi, ophiopogonis tuber, figwort root, honeysuckble and licorice root), model group and control group with 10 animals in each group. Except the control group, all the animals were models of oral ulceration, and given drugs hy squashing and infusing. The Aikuiling was given 0.5 mL for squashing with concentration of 1, 0.5, 0.25 g/mL, and 20 mL/kg for infusing; the Kouyanqing granule was given 0.5 mL for squashing and 10 mL/kg for infusing with the concentration of 0.167 g/mL; the model group was squashed with 0.5 mL normal saline, and infused 10 mL/kg normal saline; the control group was not given any intervention. The drug was given once daily for 5 continuous days. The ulceration condition was taken photograph at days 1, 3, and 5. Two hours after giving the medicine for the last time, the ulceration part was removed to make pathological section and observe the local congestion oedemardegree and the histomorphology of the ulceration. RESULTS: All the 60 animals enter into the result analysis. (1)Compared with the model group, the 20, 10, 5 g/kg doses Aikuiling groups and the KouYanQing granule group could remarkably reduce the acreage of ulcera tion at the third day [(20.30±4.6), (11.8±3.2), (8.9±6.6), (13.4±4.7), (13.0±5.8) mm^2, P 〈 0.01]. At the 5th day, there were significant differences in the acreage of ulceration among the 20 g/kg dose Aikuiling group, Kouyanqing group and the model group [(3.9±5.6), (12.2±6.2) mm^2; P 〈 0.01]; so were the 10, 5 g/kg doses Aikuiling group, Kouyanqing group and the model group [(6.1±3.0), (7.0±3.6), (6.3±3.3) mm^2; P 〈 0.05]. (2)The 20 and 10 g/kg doses of Aikuiling groups and the Kouyanqing granule group could obviously reduce the acreage of ulceration at day 3 (P 〈 0.05); at day 5, the 20 g/kg Aikuiling group lessened the ulceration degree significantly (P 〈 0.01), the 10 g/kg Aikuiling group and Kouyanqing group also lessened the ulceration degree (P 〈 0.05); the 20 g/kg Aikuiling group much reduced the ulceration compared with the Kouyanqing group (P 〈 0.05). (3)The 20 g/kg and 10 g/kg doses Aikuiling groups and the Kouyanqing granule group remarkably reduced the pathological change of the ulceration tissue (P 〈 0.01); and the 5 g/kg dose Aikuiling group only had the trend to reduce the pathological change of the ulceration tissue. CONCLUSION: Aikuiling liquid can remarkably alleviate the symptoms of oral ulceration, and obviously accelerate the ulceration healing of the guinea pigs. The effect of the 20 g/kg dose Aikuiling is better than Kouyanqing granule.
