摘要
目的:建立双氯芬酸胆碱(DAC)高效液相色谱测定方法。方法:采用C18色谱柱,以甲醇-乙腈-磷酸盐(体积比为25:25:50)为流动相,检测波长276nm,进样量20μl。24只家兔,用DAC滴眼液50μl后,取房水用甲醇处理后,取上清液进样测定。结果:本方法线性范围10~500mg/L,r=0.99981,回收率为101.3%,日内和日间RSD均〈5%。家兔用药后,房水中15min既可检测到药物,在给药后30min达到最高浓度。结论:该方法准确灵敏,精密度好,适合于眼房水中DAC的测定。
Aim: To establish an HPLC method for the determination of diclofenac acid-choline (DAC) in aqueous humor of rabbits. Methods: The concentration of DAC in aqueous humor of 24 rabbits treated with DAC eyedrop (50 μl) was determined by HPLC with diamonsil C18 as the chromatographic column, methanol-water-phosphoric acid (25 : 25 : 50)as the mobile phase, detection wavelength of 276 nm and injection volume of 20 μl. Results: The linear range was 10-500 mg/L, r =0. 999 81. The mean recovery rate was 101.3%. The intra-day and inter-day RSD were less than 5%. DAC could be determined within 15 min and reached peak concentration at 0.5 h after administration. Conclusion: The method is sensitive and accurate. And it is suitable for DAC ascertainment in aqueous humer.
