Effects of hyperbaric oxygen on brain injury and edema after head trauma in rats

摘要 目的:探讨高压氧对大鼠颅脑创伤后脑损伤和脑水肿的影响。方法:将大鼠随机分为对照组、脑创伤组和高压氧(HBO)组。采用BIM-Ⅲ型撞击机撞击大鼠右侧颅顶,复制闭合性颅脑损伤,于伤后24小时由心脏取血处死,测定脑组织含水率、伊文斯蓝(EB)含量及脑组织和血浆脂质过氧化物──丙二醛(MDA)含量。结果:(1)脑外伤组大鼠右侧脑组织含水率、脑组织及血浆MDA含量均显著高于对照组(P<0.01或P<0.05);右侧脑组织含水率及MDA含量显著高于对侧(P<0.01或P<0.05),右侧脑组织EB含量较对照组升高,但P>0.05;(2)HBO组右侧脑组织含水率及脑组织MDA含量显著低于脑外伤组(P<0.01或P<0.05),而与对照组及左侧无明显差别;血浆MDA、脑EB含量与对照组、脑外伤组及左侧均无明显差别。结论:HBO能减少脑外伤脑组织氧自由基的生成,减轻脑损伤和脑水肿。 Aim: To investlgate the effect of hyperbraric oxygen on brain injury and edema after head trauma. Methods: The rats were randomized into control(C)group,head trauma(HT)group and head trauma treated with hyperbaric oxygen(HBO) group.A model of closed craniocerebral injury by using the BIM-Ⅲ bioimpact machine to hit the rat on the right top of the head was established.The rats were then killed by drawing blood from the heart at 24 h after the trauma. The brain water content ra-tio, Evans blue and malondialdehvde (MDA)concentrations in the brain tissue and plasma were deter-mined. Results:(1) The water content ratio and MDA concentrations in the right brain tissue and plas-ma were significantly higher in HT group than that in C group(P<0. 01 or P<0. 05);the water con-tent ratio and MDA concentration in the right brain tissue were significantly higher than that in the left of the HT group(P<0.01 or P<0.05);but there was no significant difference of the concentration of Evans blue between HT and C group or the right and left brain,(2) The water content ratio and MDA of the concentration of the right brain were significantly less in HBO group than in HT group(P<0.01 or P<0.05),but there was no significant difference between HBO group and C group or the right and left brain(P>0.05);and no significant differences of the concentration of Evans blue and plasma MDA concentration were seen between HBO group and C or HT group, or between the right and left brain. Conclusion: HBO can decrease the production of the oxygen-derived free radicals in the brain after head trauma and alleviate brain inlury and edema.
