摘要
为了使人表皮生长因子(hEGF)基因在番茄中表达,构建了hEGF基因的表达载体p2300-2A11-hEGF,通过电激法转化到农杆菌菌株C58中,采用叶盘法转化番茄品系cg2002-14。子叶经发芽及生根诱导培养后得到了再生植株。PCR分析表明,hEGF基因已整合到番茄的基因组中。放射免疫法(RIA)测得重组人表皮生长因子(rhEGF)在鲜重果实中平均含量为3.85±0.86ng/g,对酒引起的小鼠急性胃溃疡有明显的预防作用,溃疡指数由52.20±23.24降至21.20±6.84,抑制率达59.39%。
To produee human epidermal growth thetor (hEGF) in transgenic mini-tomatoes, plasmids p2300-2A11-hEGF carrying the hEGF gene were eonstnaeted and transtormed via eletro-perfioration to Agrobaeterium C58, and then transteeted to tomatoes cg2001-14 via leaf disk transformation. The regenerated plantlets were obtained from the cotyledons after shoot and root inducing euhivations. The PCR analysis showed that the hEGF gene has been integrated into the mini-tomato genome. Itadioimmunoassay (RIA) showed that the mean yield of rhEGF in the fresh fruits of the transgenie mini-tomatoes amounted to 3.85 ± 0.86 ng/g frnit. Ingestion via gastric intuhation of the transgenie minitomato juice significantly protected the mice from the alcohol-induced acute peptic ulceration. The ulcer index, expressed as a degree of the stomach lesion, decreased from 52.20±23.24 to 21.20±6.84,and the inhibition rate got to 59.39%.
