摘要
[目的]建立动物模型,为化疗性静脉炎的发病机制研究以及采取有效的防护方案提供可靠的实验研究依据。[方法]将实验兔12只随机分为3组,取其双侧耳缘静脉注射诺维本低剂量组为0.4mg/kg,中剂量组为1.0mg/kg,高剂量组为1.6mg/kg同时每剂量组双耳缘静脉分别推注10mL和20mL容量的药液(药液采用不同容量的生理盐水稀释)。分别观察静脉炎发生的时间,并在实验后48h及第7天进行病理组织切片比较。[结果]高剂量组实验兔病死率高,静脉炎症重,中剂量组无死亡,静脉炎症中度,低剂量组静脉炎症轻,病理特征不明显,适合作为静脉炎模型在剂量相同的条件下,药液稀释容量不同,兔耳静脉的炎症反应不同,20mL组病理损伤重于10mL组。[结论]采用兔耳静脉双侧注射中剂量诺维本(1.0mg/kg),药液稀释至20mL静脉推注能够较好地建立化疗性静脉损伤的动物模型。
Objective: To establish animal models, to provide an experimental study basis for both the pathogenesis study and taking effective prevention scheme on chemotherapeutic phlebitis. Method: A total of 12 rabbits were divided into three groups, and were injected Navelbinc into auricular vein of bilateral ears with a dose of 0.4 mg/kg in low dose group, 1.0 mg/kg in moderate dose group and 1.6 mg/kg in high dose group and diluted with 10 mL and 20 mL normal safine respectively. Then the time of phlebitis developed in all rabbits were observed, and histopathologic slides of animal modes at 48 hours and 7 days after experiment were performed and compared. Result: The fatality of experimental rabbits was high in high dose group and phlebitis was severe. In contrast, rabbits in low dose group developed mild phlebitis. None death was found in moderate dose group rabbits and phlebitis was moderate as well, which is suitable to be prepared as phlebitis model. Under the same dose, phlebitis reactions of auricular vein in rabbits were different as it diluted in different amounts of normal saline. The pathological changes in 20 mL normal saline dilution group were more severe than that of 10 mL dilution group. So the dilution dose of establishing phlebitis model was suggested to be 20 mL normal saline. Conclusion: It would be feasible to establish an auricular vein phlebitis model of rabbits by intravenously infusion of Navelbine at a dosage of 1.0 mg/kg and diluted with 20 mL normal saline via bilateral auricular vein.
