丙烯酰胺致大鼠睾丸生殖细胞DNA损伤与P53蛋白的表达 被引量:3

DNA DAMAGE AND P53 PROTEIN EXPRESSION OF TESTES CELLS OF MICE AFTER ACRYLAMIDE EXPOSURE

摘要 目的:研究丙烯酰胺慢性染毒后睾丸生殖细胞DNA损伤与p53蛋白表达的关系,为丙烯酰胺生殖毒性及其修复机制研究提供基础数据。方法:昆明种小鼠经丙烯酰胺(20、40、60 mg/kg体重)慢性染毒8周后,单细胞凝胶电泳研究睾丸生殖细胞的DNA损伤;免疫组化采用SABC法。结果:丙烯酰胺染毒8周后的各组受损细胞增加,形似彗星;40、60 mg/kg组的细胞拖尾率及尾长显著高于阴性对照(P<0.05);60 mg/kg组的细胞中呈现较多棕黄色颗粒,p53表达在20、40、60 mg/kg各组的阳性率分别为18.1%、21.2%和29.8%,与阴性对照比较60 mg/kg组阳性率有显著差异(P<0.05)。结论:丙烯酰胺有明确的DNA损伤作用,p53蛋白参与了损伤细胞的修复或凋亡途径。 Objective: To investigate the relationship between DNA damage and p53 expression after acrylamide exposure of mice. Methods: Single cell gel electrophoresis was used to determine the DNA damage. P53 expression was probed with immunohlstochemical assay of SABC. Results: Comet tails were observed in the positive controls and all the acrylamide exposure groups. The comet cell rate and tail length in 40 mg/kg and 60 mg/kg b. w. groups were significantly higher than that of negative group (P〈 0.05). p53 expression was significantly higher in 60 mg/kg b. w. groups than that of negative control (P〈0.05). Conclusion: It suggested that acrylamide took a definite DNA damage to testes cells from mice. P53 played a role in the apoptosis of testis cells after acrylamide exposure.
