摘要
本文对与乙型肝炎病毒外周蛋白preS1特异结合的B细胞来源的Wa细胞膜受体样蛋白进行了研究,用麦芽糖结合蛋白-preS1-直链淀粉树脂亲和层析柱分离得到了与preS1特异性结合的Wa细胞膜受体样蛋白质,抗麦芽糖结合蛋白-preS1的单克隆抗体F35.25结合Westernblot证实确系膜蛋白与preS1的特异性结合,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳和聚丙烯酰胺凝胶电泳结果显示,Wa细胞膜上存在有三种不同的受体样蛋白,分子量分别为60000,56000和48000.这表明在非肝细胞膜中也可能存在乙型肝炎病毒的特异性受体和直接感染途径。
The receptor-like proteins of Wa cells bound to preS1 of hepatitis B virus were isolated by using maltose binding protein (MBP)-preS1-amylose affinity chromatography and identified by Western blot using a monoclonal antibody,F35.25, which was specific for MBP-preS1.The mass molecular weights of the receptor-like proteins isolated were approximately 60000,56000 and 48000, respectively. The results indicated that specific receptor for preS1 of hepatitis B virus seem to be existed in some extrahepatic cell lines and direct infect pathway may be exist.
