RP-HPLC法测定润肠丸中阿魏酸及5种蒽醌衍生物的含量 被引量:3

摘要 目的:建立润肠丸中阿魏酸和蒽醌衍生物芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚6种成分的高效液相色谱测定方法。方法:Hypersil ODS-2 C_(18)柱(5 μm,4.6 mm×250 mm),Kromasil ODS 保护柱(5 μm,4.6 mm×10 mm)流动相:甲醇-1%醋酸,梯度洗脱,流速:0.8及1.0 mL·min^(-1),二极管阵列检测器(PDA),检测波长:阿魏酸323 nm,大黄蒽醌衍生物430nm,柱温:25℃。结果:可在2个波长下同时测定6种化合物的含量。阿魏酸、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的线性范围分别为0.04~1.41,0.03~2.00,0.03~2.05,0.02~1.45,0.04~2.11,0.03~1.74 μg。平均回收率分别为104.5%,101.0%,101.0%,100.9%,95.5%,101.1%;RSD 分别为0.14%,0.31%,1.2%,0.28%,0.21%,1.3%。结论:该方法灵敏,准确,是检测润肠丸中阿魏酸和5种蒽醌衍生物较理想的方法,为该制剂的质量标准提升提供了依据。 Objective: To establish an HPLC method for the simultaneous determination of the contents of ferulic acid, aloe emodin,rheins,emodin,chrysophanol and physcion in Runchang pills. Methods:The separation was performed on Hypersil ODS -2 C18 column(5 μm,4. 6 mm ×250mm)and Kromasil ODS guard column(5 μm,4. 6 mm× 10 mm)with a mobile phase of methanol- 1% acetic acid gradient elution,and the flow rate were 0. 8 and 1.0 mL · min^-1. Photodiode Army(PDA) detector was used and the wavelength of detector for ferulic acid and anthraquinones ramification were set at 323 nm and 430 nm respectively, and 25 ℃ for column. Results :The contents of 6 compounds were simultaneously detected at two wavelengths. The calibration curves showed good linearity in the range of 0. 04 - 1.41 μg(ferulic acid) ,0. 03 - 2.00 μg(aloe emodin) ,0. 03 -2.05 μg(rhein),0.02 -1.45 μg(emodin),0.04 -2. 11 μg(chrysophanol) and 0.03 - 1.74 μg(physcion) ,and the average recoveries of the method were 104. 5%, 101.0%, 101.0%, 100. 9% ,95. 5% and 101.1% respectively with corresponding RSD of 0. 14% ,0. 31%, 1.2% ,0. 28% ,0. 21% and 1.3%. Conclusion: The method is accurate and sensitive. The method is ideal for controlling quality of the preparation.
