摘要
目的获得具有溶解纤维蛋白能力的中草药。方法选择22种具有活血化瘀功能的中草药,分别进行蛋白质的提取,对蛋白质提取物进行纤维蛋白溶解活性的鉴定。结果10种中草药具有溶解纤维蛋白的能力,作用强弱顺序为肉豆蔻、莪术、益母草、茜草、延胡索、皂角刺、祖师麻、石蒜、九里香、姜黄,其中肉豆蔻纤维蛋白溶解圈最大而且透明。对出现溶圈的蛋白质提取物进行浓度测定,根据尿激酶标准曲线,计算其溶解纤维蛋白的比活力。益母草、延胡索、肉豆蔻、茜草的比活力相对较高,分别为582.43,543.18,480.20和264.52 U/mg。结论建立了筛选具有纤溶酶活性的中草药的方法,并筛选出4种纤溶酶活力相对较高的中草药。
Purpose To obtain some Chinese herbal medicines with fibrinolytic enzyme. Methods Proteins were extracted from 22 drugs which were used to activate blood circulation and to remove blood stasis, and were observed in fibrin plate. Results The crude proteins of 10 drugs produced fibrinolytic activity in fibrin plate. They were Nutmeg ( Myristica fragrans ) , Ezhu ( Curcuma phaeocaulis ), Motherwort ( Leonurus japonicus ), Madder( Rubia cordifolia ) , Yanhusuo ( Corydalis yanhusuo ), Chinese Honeyloeust Spine ( Spina Gleditsiae ) , Cirald Daphne Bark ( Daphne giraldii ), Shorttube Lycoris ( Lycoris radiate ), Folium Et Cacumen Murrayae (Murraya paniculata), and Curcumae( Curcuma longa) in high to low order. Among them, Nutmeg showed the biggest and most transparent fibrinolytie zone. The concentration of proteins was measured by Bradford. Then, specific activity was calculated according to the standard value of urokinase plasminogen activator. It showed that Motherwort, Yanhusuo, Nutmeg and Madder had higher specific activity than others. They were 582.43, 543. 18, 480.20 and 264.52 U/nag respectively. Conclusion The method of screening Chinese Herbal Medicines with fibrinolytie enzyme was established. And there were four drugs which had stronger fibrinolytic enzyme than others.
