摘要
[目的]探讨焦炉工HSP70基因多态性和外周血淋巴细胞DNA损伤的关联。[方法]选择某焦化厂234名焦炉作业工人和130名无多环芳烃(PAHs)及其他毒物暴露者,采集外周静脉血,分离淋巴细胞,使用单细胞凝胶电泳实验评价DNA损伤;应用多聚酶链反应-限制片段长度多态性(PCR-RFLP)技术检测热休克蛋白70(HSP70)基因多态性;运用PHASE2.0.2遗传分析软件计算单倍型。[结果]焦炉工外周血淋巴细胞的Olive尾矩(OliveTailMoment,OTM)高于对照组(对数转换后分别为1.29±1.07、0.56±0.99),差异有极显著性(P<0.01)。焦炉工中,HSP70-1基因190位点C/C基因型携带者外周血淋巴细胞的OTM为2.19±0.65,高于G/C及G/G基因型携带者(分别为1.30±1.03、1.23±1.10),差异有极显著性(P<0.01);HSP70-2基因1267位点A/A、A/G及G/G基因型携带者OTM分别为1.23±1.02、1.27±1.06和1.42±1.18,三者间差异无显著性(P>0.05);HSP70-hom基因2437位点T/T、T/C及C/C基因型携带者OTM分别为1.32±1.09、1.25±1.01和0.83±0.96,三者间差异也无显著性(P>0.05)。焦炉工HSP70-hom基因2437位点、HSP70-1基因190位点和HSP70-2基因1267位点组成的单倍型对中,TCG/TCG单倍型对携带者DNA损伤程度较重(OTM为2.04±0.59),显著高于单倍型对中含一个TCG单倍型者(OTM为1.24±1.14)及不含TCG单倍型者(OTM为1.30±1.01),P<0.05。[结论]焦炉工外周血淋巴细胞HSP70-1基因190位点C/C基因型与TCG单倍型和DNA严重损伤有关联。
[ Objective ] To study the relationship between HSPTO gene polymorphisms and DNA damage in lymphocytes of coke oven workers. [ Methods ] 234 coke oven workers and 130 employees who had no exposure history to PAHs and other toxicants were selected as controls. Lymphocytes were separated from their peripheral venous blood. Single cell gel electrophoresis was used to evaluate DNA damage, and polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP ) was used to analyze HSP70 gene polymorphisms. Using PHASE 2.0.2 software to calculate haplotype. [ Results ] The lymphocytes olive tail moment( OTM )of coke oven workers was significantly higher than that of controls ( 1.29 ± 1.07 vs 0.56 ± 0.99, P 〈 0.01 ). Among coke oven workers, the OTM of those with C/C genotype at HSP70-1 gene 190 was 2.19 ± 0.65, which was significantly higher than that of those with G/C and G/G genotype ( 1.30 ± 1.03 and 1.23 ± 1.10 respectively, P 〈 0.01 ); The OTM had no significant difference among those with A/A, A/G and G/G genotype at HSP70-2 gene 1267 site( 1.23 ± 1.02, 1.27 ± 1.06 and 1.42 ± 1.18, P 〉 0.05 ). And the OTM also has no significant difference among coke oven workers with T/T, T/C and C/C genotype at HSP70-hom gene 2437 site( 1.32 ± 1.09, 1.25 ± 1.01 and 0.83 ± 0.96, P 〉 0.05 ). The coke oven workers with TCG/TCG haplotype pair of HSP70-hom, HSP70-1 and HSP70-2 gene had more severe DNA damage ( 2.04 ± 0.59 ) than those with only one TCG haplotype( 1.24 ± 1.14 )and those without TCG haplotype ( 1.30 ± 1.01 ), with significant difference( P 〈 0.05 ). [ Conclusion ] The gene polymorphism at HSP70-1 gene 190 site and TCG haplotype had association with severe DNA damage.
