摘要
目的:观察当归补血汤对耳蜗干细胞体外诱导分化的影响,并探讨浓度对其的影响。方法:分离新生大鼠Corti器的耳蜗干细胞进行细胞培养,在相差显微镜下观察细胞形态及生长状况,用Nestin免疫荧光法进行耳蜗干细胞鉴定。采用不同剂量当归补血汤进行诱导分化后,采用MyosinⅦA、P27^(KIP1)免疫荧光方法,鉴定内耳毛细胞和支持细胞,并进行细胞计数。结果:当归补血汤细胞诱导分化的内耳毛细胞和内耳支持细胞数量明显较对照组多,其中剂量为0.5 g/ml组向内耳毛细胞的分化比例最高。结论:当归补血汤可以提高耳蜗干细胞体外分化为内耳毛细胞和支持细胞的数量。其中剂量0.5 g/ml组向内耳毛细胞的分化数量较高。
Objective: To investigate the probabilities of corti tissues stern cells from cochlea tissues of neonatal rat differentiating into hair cells and supporting cells by astragalus membranaceus and angelica sinensis. Methods: The cochlea stem cells were cultured by using the method of serum-free incubation and single-cell cloning technique and the growth characteristic of the stem cells was observed. The expression of nestin was used to determine the stern cell characters. Then the cochlea stern cells were cultured at different concentrations in the astragalus membranaceus and angelica sinensis. Myosin ⅦA and P27^KIP1 immunofluorescence was used to detect the hair cells and supporting cells. Results: The number of the hair cells and the supporting cells cultured in the astragalus membranaceus and angelica sinensis groups was higher than that in the control group. And the number of hair cells in 0.5 g/ml astragalus membranaceus and angelica sinensis was the highest after indudced differentiation, but the number of the supporting cells was not related to the concentration of the astragalus memhranaceus and angelica sinensis. Conclusion: Astragalus membranaceus and angelica sinensis can increase the number of Myosin Ⅶ A(+) hair cells and supporting cells. The differentiation number of the hair cells in the 0.5 g/ml group is higher than that in the astragalus membranaceus and angelica sinensis groups.
