摘要
目的分析和比较PCR方法与血清学方法对三种病原体的检测应用结果。方法用已建立的检测弓形虫、布氏杆菌、轮状病毒核酸的PCR方法分别对ICR小鼠的肝组织、Beagle犬的粪便和阴道拭子,猕猴的粪便进行了相关病原体的核酸检测。同时用血清学抗体检测方法对ICR小鼠、Beagle及猕猴血清进行了相关病原体抗体检测。结果核酸检测结果和用血清学方法所得的检测结果,以及用不同血清学方法所得的检测结果相一致。结论作者所建立的三种病原体的PCR检测方法具有较好的特异性、敏感性和操作简便的优点,可以和血清学方法一起使用于实验动物的微生物质量检测,并弥补血清学方法的不足。
Objective Comparative the results of PCR and serology methods for detecting of three different pathogenes. Methods The nucleic acid of Toxoplasma gondii, Brucella and Rotavirus extracted from the liver of ICR mice, stool speciments and vagina specimen of Beagle, stool specimen of Macaca mulata respectively was amplified by NT-PCR, PCR and RT-PCR. The serum antibody of Toxoplasma gondii, Brucella and Rotavirus from ICR mice, Beagle and Macaca mulata respectively was detected by serology methods. Results The results of NT-PCR, PCR, RT-PCR were accorded with serology methods.Conclusion The developed NT-PCR, PCR, RT-PCR assay was a rapid, sensitive and specific method, it can be used in monitoring pathogen.
