摘要
目的:探讨吗啡戒断后不同时段焦虑小鼠下丘脑β-内啡肽(β-EP)、5-羟色胺(5-HT)及其代谢产物5-羟吲哚乙酸(5-HIAA)含量的变化,以及电针的作用效应。方法:按照逐日递增原则给予小鼠皮下注射吗啡,以纳洛酮催瘾,建立吗啡戒断后焦虑动物模型。电针“三阴交”穴,采用放射免疫法检测下丘脑β-EP的水平,荧光法检测5-HT和5-HIAA含量。结果:1.电针可明显提高吗啡戒断3天和5 d后焦虑小鼠下丘脑β-EP水平(P<0.05,或P<0.01);2.电针同时能降低吗啡戒断3 d和5 d后下丘脑5-HT含量(P<0.05)、降低5 d后5-HIAA(P<0.05)含量。结论:电针抗吗啡戒断后焦虑效应可能与其通过提高脑内内源性β-EP水平,从而下调焦虑小鼠5-HT的合成、释放及其代谢有关。
Objective: To explore the effect on the Hypothalamus of β-EP, 5-HT and 5-HIAA content in the anxiety model mice after morphine withdraw different time by electro-acupuncture. Methods: The anxiety mice model was formed based on the principle of morphine increase progressively day by day, in which after the mice were induced into morphine dependence, naloxone was given. The model mice were treated with electro-acupuncture on acupoint "Sanyinjiao" (SP6). The contents of β-EP, 5-HT and 5-HLAA in the mice' s hypothalarnus were measured, respectively by the radia- tion immunoassay and fluorescence spectrophotometry. Results: The contents of β-EP in the electro-acupuncture treatment group was obviously elevated compared with model group in after 3th and 5th days morphine withdraw (P 〈 0.05, orP 〈 0.01 ) ; Simultaneous the contents of 5-HT in the electro-acupuncture treatment group was obviously decreased compared with model group in after morphine withdraw 3th and 5th day (P〈 0.05), and the contents of 5-HLAA was obviously decreased compared with model group in after morphine withdraw on 5th day (P 〈 0.05). Discussion: There is anti-anxiety effect of electro-acupuncture in the anxiety model mice from morphine withdraw which is possible mechanism by modulated with increasing the contents of endogenous opioid peptide and down regulation the synthesis , release and metabolism of 5-HT in the brain.
