摘要
目的观察胰岛素抵抗(IR)大鼠大网膜脂肪细胞过氧化物酶体增生物激活受体γ(PPAR-γ)mRNA的表达水平以及罗格列酮对PPAR-γmRNA表达水平的影响。方法雄性Wistar大鼠随机分为空白组、模型组、罗格列酮组,正常组喂以普通饲料,模型组与治疗组注射小剂量STZ并喂以高热量饲料,常规测定各组大鼠治疗前后的空腹血糖(FPG)和治疗后各组大鼠的空腹血清胰岛素水平(FIns),计算胰岛素敏感性指数(ISI),测定各组血脂水平,提取大网膜脂肪组织总RNA,采用逆转录PCR技术扩增PPAR-γ基因片段,检测其表达水平。结果模型组PPAR-γmRNA表达比正常组明显减少(P<0.01),与模型组比较,罗格列酮组PPAR-γmRNA表达显著增加(P<0.01)。结论R大鼠脂肪细胞PPAR-γmRNA表达减少,而罗格列酮能有效提高PPAR-γmRNA的表达。
Objective To investigate the difference of peroxisome proliferator-activated receptor--γ(PPAR-T)mRNA's expression in greater omental adipose in insulin resistance(IR) rats and Rosiglitazone Tablets'effect on it. Methods Male wistar rats were randomly divided into 3 groups (normal group, model group, Rosiglitazone Tablets treating group). The rats of normal group were fed with common food while the others with high calorie food and injected with low dose of STZ. Fasting plasma glucose(FPG) level, fasting insulin (Fins) level and blood fat of all rats were measured by routine methods,and insulin sensitivity index(ISI) was calculated accordingly. At the end of experimental treatment, the total mRNAs of greater omental adipose tissues were extracted and the genie fragments of PPAR-γwere amplificated in order to detect their expression levels. Results Rosiglitazone tablets could reduce FPG and Fins level in model rats and enhance ISI effectively.The PPAR-γmRNAs were expressed less obviously in the rats of model group than in normal group (P〈0.01). The expressional level was significantly higher in Rosiglitszone tablets treating group than model group (P〈0.01). Conclusion The PPAB-γmBNAs are expressed less obviously in IR rats,and Rosiglitazone maleate tablets can enhance its genie expressional level effectively.
