艾炷灸贴预处理对佐剂性关节炎大鼠早期和继发期血清白细胞介素-6、睾酮、生长激素及皮质醇的影响 被引量:6

Influence of Acupoint-application and Moxibustion Pretreatment on Serum IL-6,T,GH and CORT Levels in Rats with Adjuvant Arthritis at the Early and Secondary Stages

摘要 目的:探讨艾炷灸贴预处理对佐剂性关节炎(AA)大鼠早期与继发期血清炎性因子和激素的影响及保护机制。方法:将40只Wistar大鼠随机分为正常组、模型早期组和继发组、预灸贴早期组和继发组。于造模前用中药膜剂贴敷大鼠“大椎”穴后加艾炷灸,采用弗氏完全佐剂制备大鼠模型,用放射免疫法测定各组大鼠血清白细胞介素-6(IL-6)、睾酮(T)、生长激素(GH)及皮质醇(CORT)的含量。结果:血清IL-6和GH含量造模后均明显升高(P<0.01);与同期模型组相比,预灸贴继发组均明显下降(P<0.05)。血清T含量造模后早期组明显降低(P<0.05),继发组变化不大;预灸贴各组较同期模型组均显著升高(P<0.01,0.05)。血清CORT含量造模后显著升高(均P<0.01),而预灸贴继发组与正常组比无明显差异(P>0.05)。结论:艾炷灸贴预处理可减轻随后佐剂性关节炎大鼠原发足局部病理损伤,降低血清炎性因子IL-6的水平,对血清T、GH及CORT等内分泌激素的紊乱状态具有良性调节作用。 Objective: To observe the effects of acupoint-application and moxibustion pretreatment on serum inflammatorycytokine and hormones of rats with adjuvant arthritis at early and secondary stages. Methods: Forty Wistar rats were randomly divided into normal, early-stage model, secondary-stage model groups, early-stage pretreatment and secondary-stage pretreatment groups. Arthritis model was established by injecting Freund's complete adjuvant (FCA, 0. 1 mL) into the rat's right hindpaw. After removing the hair and before injecting FCA respectively, a plaster made of Herba Epimedii, etc. and an ignited moxa cone (placed on the plaster) were applied to "Dazhui" (GV 14), once every other day, 8 times altogether. Rats of the 4 earlystage and secondary-stage groups were killed on the 3^rd day and 16^th day after injection of FCA respectively, and those of normal group also killed for collecting blood samples. The contents of serum IL-6, testosterone (T), growth hormone (GH) and cortisol (CORT) were assayed respectively with radioimmunoassay according to the instructions of reagent kits. Results: Compared with normal group, the contents of serum IL-6, GH and CORT of early-stage-model and secondary-stage-model groups increased significantly (P〈0.01), and those of early-stage pretreatment groups, and serum T and GH of secondary-stage pretreatment groups also increased remarkably ( P〈0.05, 0.01 ) ; but T level of early-stage model group decreased significantly ( P〈O. 05) ; while in comparison with the corre-sponding model groups, the contents of T in early-stage and secondary-stage pretreatment groups increased considerably (P〈0.05,0.01 ) ; and those of serum IL-6 and GH lowered markedly (P〈0.05) ; no significant differences were found among the 2 model and 2 pretreatment groups (P〉0.05). Conclusion: Preconditioning of acupoint-application and moxibustion at GV 14 can effectively suppress arthritis induced increase of serum IL-6 and GH levels and decrease of T to modulate inflammatory cytokine and endocrine hormones, playing a protective role in adjuvant arthritis rats.
