4′,5′,7′-三羟基异黄酮对肺癌细胞的抑制作用及其机制

摘要 目的:探讨4′,5′,7'三羟基异黄酮对肺癌细胞的抑制作用及其相关的机制。方法:选择人肺腺癌细胞株A549为研究对象,分成3组:Genistein处理组、无水乙醇对照组和正常对照组。各组细胞应用MTT方法检测细胞的抑制率,应用流式细胞仪检测细胞的周期,并计算增殖指数,并应用流式细胞仪检测细胞线粒体的膜电位的变化。结果:应用Genistein处理后细胞增殖受到明显抑制,表现为MTT吸收值降低,PI下降;同时ΔΨm下降的细胞百分率增加。结论:Genistein可以抑制肺癌细胞的增殖,其机制是通过改变细胞线粒体膜电位而促进肿瘤细胞的凋亡。 Objective To investigate the inhibitory effect and correlative mechanism of genistein on pulmonary carcinoma cell. Method We selected the human adenocarcinoma of lung ceil line A549 as the study object, and divided them into three groups as follows: genistein treated group, absolute alcohol control group and normal control group. The suppression rate was detected by MTT, the cell cycle and transmembrane potential of mitochondrium(△Ψm) were detected by flow cytometry and the proliferation indexes(PI) was calculated. Results The cells treated with genistein were suppressed obviously and showed descent in absorption value of MTT. PI degression and increasing percentage of cells with decreasing △Ψm were found. Conclusion Genistein can suppress the proliferation of adenocarcinoma of lung cell line A549 and the mechanism is the apoptosis induced by the changing △Ψm.
