摘要
目的比较的小鼠CD90+Lin-、C34+Lin-、CD117+Lin-和Sca-1+Lin-骨髓细胞的肝干细胞潜能。方法供体为纯系BALB/C雄性小鼠,应用免疫磁珠分选法获取CD90+Lin-、CD34+Lin-、CD117+Lin-和Sca-1+Lin-骨髓细胞。受体为35Gy全肝照射处理后的同龄同系BALB/C雌性小鼠,A、B、C、D组小鼠分别接受CD90+Lin-、CD34+Lin-、CD117+Lin-、Sca-1+Lin-骨髓细胞移植。移植后30天活杀小鼠,取肝进行Y染色体原位分子杂交与白蛋白免疫组织化学联合检测。结果各组小鼠肝组织中都可以检测到Y染色体和白蛋白均呈阳性的双阳性细胞,C组的双阳性细胞数量显著多于其它各组。结论CD90+Lin-、CD34+Lin-、CD117+Lin-、Sca-1+Lin-骨髓细胞都具有肝干细胞潜能,其中CD117+Lin-骨髓细胞的肝干细胞潜能最大、形成肝细胞的能力量强。
Objective To compare the hepatic stem cell potential of CD90 + Lin-, CD34 + Lin-, CD117 + Lin-and Sca-1 + Lin- bone marrow cells in mice. Methods CD90 + Lin-,CD34 + Lin-,CD117 + Lin-and Sca-1 + Limbone marrow cells were isolated and purified by magnetic-activated cell sorting from BALB/C male donor mice. The recipients in group A, B, C, D were age-matched BALD/ C syngeneic female mice with 35-Gy total liver irradiation and received CD90 + Lin,CD34 + Lin,CD117 + Lin,and Sca-1 + L/n, bone marrow cells respctively. The mice in all groups were sacrificed 1 moth after the transplantation. The liver were examined by situ hybridization for Y-chromosome and immunohistochemical staining for albumin. Results The Y-chromosome and positive and albumin posi- tive cells were detected in the recipient's liver of all groups. The number of the two positive cells in group C was significantly morw than those in group A, B, C, D. Conclusion CDg0 + Lin-, CD34 + Lin-, CD 117 + Lin-, and Sea-1 + Lin-bone marrow cells have the potential of hepatic stem cells, but the best cell in differentiation into hepatocytes in the liver after transplantation was CD117 + Lin-Lin-bone marrow cell.
