利用组织芯片研究Fascin在食管癌中的表达 被引量:10

摘要 目的:本研究利用组织芯片技术高通量地研究Fascin蛋白在食管癌中的表达。方法:采用56例食管癌病人的病理蜡块制作组织芯片,应用免疫组化的方法检测Fascin在食管癌组织中的表达情况。同时对8例食管癌标本的冰冻切片显微切割“纯”癌和非癌组织,进行蛋白印迹研究。结果:在55例可提供信息的病例中,总体上72.7%的肿瘤中为阳性染色。其中高分化阳性率为95.2%,中分化为75%,低分化为35.7%,Fascin表达与食管癌分化有关(P<0.001)。蛋白印迹结果也证实在分化较好的食管癌中Fascin为高表达。结论:利用组织芯片可以应用于回顾性Fascin表达的研究,研究中发现食管癌中Fascin的表达与组织分化有关。该试验方法简便,易于在临床推广。 Objective:Fascin is a prognostic factor for Esophageal Scluamous Cell Carcinoma (ESCC). The aim of this study was to explore the expression of Fascin in ESCC by tissue array analysis. Methods: Tissues from 56 ESCC patients were arranged into one wax block by tissue arrayer. Immunohistochernistry was applied to evaluate the expression of Fascin. Mean while, 8 paired tumor and non-cancerous normal tissues were cryosectioned and "pure" cancer and normal tissue were separated by laser captured rnicrodissection. Expression of Fascin in these captured tissues was validated by western blot. Results: In 55 informative cases, overall 72. 7% tumor was Fascin positive. With regard to tumor differentiation, Fascin positive rate was 95.2% in well differentiated tumor, while 75% in intermediate differentiated tumor and 35.7% in lower differentiated tumor. And the expression related to tumor differentiation (P〈0. 001). Results of western-blot proved high expression of Fascin in well to intermediate differentiated tumors. Conclusion:Tissue array can be applied to retro-respectively evaluate the expression of protein Fascin. In this study, Fascin expression was found tumor differentiation related in ESCC.
